坏死性小肠结肠炎诱导全身炎症反应综合征动物模型血浆细胞因子表达变化 被引量:6

The expression of plasma cytokine in systemic inflammatory response syndrome model rats induced with necrotizing enterocolitis

摘要 目的 探讨坏死性小肠结肠炎(NEC)诱导的全身炎症反应综合征(SIRS)动物模型血浆细胞因子水平.方法 40只新生Wistar大鼠,日龄36 ~48 h,随机分为对照组10只及NEC模型组30只.模型组大鼠采用低温缺氧、脂多糖10 mg/kg灌胃制备NEC模型,鼠乳代用品人工喂养,对照组采用生理盐水灌胃,不进行低温和缺氧.实验结束后处死动物,模型组根据血液白细胞总数及呼吸改变确定是否发生SIRS,并进一步分为未发生SIRS组和发生SIRS组.采用液相芯片技术比较血浆白细胞介素(IL)-1β、肿瘤坏死因子(TNF)-α、IL-6和IL-10的含量.结果 与对照组比较,模型组血浆细胞因子IL-1β、TNF-α、IL-6、IL-10水平均升高,且发生SIRS组细胞因子水平高于未发生SIRS组(P均<0.05).对照组、未发生SIRS组和发生SIRS组各细胞因子水平分别为:IL-1β(0.100±0.016)、(3.187 ±0.525)、(4.182±0.552)pg/ml; TNF-α (0.027 ±0.008)、(0.854±0.101)、(1.077±0.133)pg/ml;IL-6(0.033±0.006)、(0.984±0.102)、(2.039±0.483) pg/ml; IL-10 (0.056±0.015)、(1.978±0.096)、(2.810±0.435) pg/ml.结论 NEC诱导的SIRS新生大鼠模型中,血浆IL-1β、TNF-α、IL-6、IL-10含量升高,提示细胞因子之间的相互调节可能影响SIRS的进程. Objective The role of cytokines in systemic inflammatory response syndrome (SIRS) was evaluated in rats induced with a NEC model. Methods 40 Wistar newborn rats (age 36 48 h) were randomly assigned into control group (u = 10) and experiment group (n = 30). The rats in experiment group were formula fed and were exposed to hypoxic and hypothermic conditions, then received LPS (10 mg/kg) intragastric irrigation. Control group rats were not exposed to hypoxic or hypothermic conditions and were irrigated with normal saline. SIRS was diagnosed by blood leukocyte count and respiratory rate of the neonatal rats in the treatment group. Experiment group was subdivided into rats who did not develop SIRS and those who did. Blood was collected immediately after the animals were killed, then blood samples were centrifuged. The plasma was collected and stored at -80℃. The level of IL-1β, TNF-β, IL-6 and IL-10 in plasma were detected by liquid chip. Results Compared with control group, The levels of IL-1β, TNF-α, IL-6 and IL-10 in plasma in experiment group were increased and group that develop SIRS was higher than in group that did not ( both P 〈 0.05 ). The levels of IL-1β(pg/ml) in control group, group that did not develop SIRS and group that did were respectively(0. 100±0. 016), (3. 187±0. 525) and (4. 182±0. 552). The levels of TNF-α(pg/ml)in the three groups were respectively (0. 027 ±0. 008), (0. 854 ±0. 101 ), and ( 1. 077 :tO. 133). The levels of IL-6 (pg/ml) in the three groups were respectively (0. 033 ±0. 006), (0. 984 ± 0. 102) and (2. 039 ± 0. 483). The levels of IL-10 (pg/ml) in the three groups were respectively (0. 056 ±0. 015), (1. 978±0.096) and (2.81 ±0.435). Conclusions The levels of IL-1β, TNF-α, IL-6 and IL-10 were increased in SIRS/NEC model newborn rats, suggesting that cytokine network interactions may affect the process of SIRS.
