白细胞介素2和白细胞介素6对肝癌细胞表达血管内皮生长因子B的调节 被引量:3

摘要 目的检测分析白细胞介素2(interleukin-2,IL-2)和白细胞介素6(interleukin-6,IL-6)刺激肝癌细胞后血管内皮生长因子B(vascular endothelial growth factor B,VEGF-B)表达的变化。方法常规培养肝癌细胞株SMMC-7721和BEL-7402,以IL-2或IL-6分别刺激细胞株后用逆转录-聚合酶链式反应(reverse transcription polymerase chain reaction,RT-PCR)技术分析其VEGF-B基因的表达。结果 IL-2刺激肝癌细胞株SMMC-7721和BEL-7402后,VEGF-B mRNA表达逐渐减少;而IL-6刺激肝癌细胞株SMMC-7721后VEGF-B mRNA的表达逐渐增加,但刺激肝癌细胞株BEL-7402后VEGF-B mRNA的表达无明显变化。结论 IL-2抑制肝癌细胞VEGF-B mRNA的表达,提示IL-2可抑制肝癌细胞的血行转移;IL-6促进部分肝癌细胞VEGF-B mRNA的表达,但对肝癌细胞生物学特性的调节有待于进一步深入研究。 Objective To explore interleukin-2 (IL-2) and intedeukin-6 (IL-6) regulation to the expression of vascular endothelial growth factor B (VEGF-B) in cultured human liver cancer cell lines. Methods Reverse transcription polymerase chain reaction(RT-PCR) was used to analyze VEGF-B mRNA expression in two cultured human liver cancer cell lines( SMMC-7721 and BEL-7402) under the stimulation with IL-2 or IL-6. Results RT-PCR analysis revealed the level of VEGF-B gene was decreased in SMMC-7721 and BEL-7402 cells by being stimulated with IL-2. IL-6 stimulated VEGF-B mRNA expression of SMMC-7721 cell, the effect was consistency-dependent, but had no effect on VEGF- B mRNA expression of BEL-7402 cell. Conclusion IL-2 inhibits blood metastasis of liver cancer cell by inhibiting expression of VEGF-B; IL-6 increases the expression of VEGF-B mRNA in some liver cancer cell, but the effect on biological characteristics of liver cancer cell needs further study.
