睾丸性不育症间质细胞增殖变化与Livin、Bim蛋白表达的关系

摘要 目的研究睾丸性不育症中睾丸间质细胞增殖变化与相关基因Livin、Bim的关系,为睾丸间质细胞增殖失衡的基因调控机制提供依据。方法应用免疫组化S-P法检测睾丸间质细胞Ki-67和Livin、Bim蛋白的表达,以Ki-67计数作为增殖指数(PI)。结果在58例睾丸性不育症间质细胞中,PI和Livin、Bim蛋白阳性表达率分别为(0.57±0.08)、36.2%、69.0%;在11例正常睾丸组织间质细胞中,分别为(0.78±0.15)、54.5%、36.4%,组间比较差异有统计学意义(P<0.05)。Livin阳性组PI高于阴性组(P<0.05),呈正相关;Bim阳性组PI低于阴性组(P<0.05),呈负相关。结论睾丸性不育症中睾丸间质细胞增殖失衡与Livin、Bim基因异常表达有关,从而间接影响精子形成。 Objective Study on the relationship between leydig cell prohferation changes and related genes in testicular infertility, to provide basis for the gene regulation mechanism of imbalance of Leydig cell proliferation. Methods The expression of ki - 67, livin and bim protein were detected by using the method of S - P immunohiswchemistry, the count of Ki - 67 was used as proliferation index (PI). Results PI, positive rate of Livin and Bim protein were 0.57 + 0.08, 36.2% and 69.0% respectively in the testicular infertility leydig ceils ( n = 58), and those were O. 78 _+ 0. 15, 54.5% and 36.4% respectively in normal testicular tissue leydig ceils (n = 11 ). The difference between the two groups was significant ( P 〈 0. 05 ). The PI in normal testicular tissue leydig cells was significantly higher than that in testicular infertility leydig cells, and positively correlated to the expression of Livin, negatively correlated to the expression of Bim(P 〈 O. 05). Conclusions The Leydig cell proliferation imbalance in testicular infertility was correlated to abnormal expression of Livin and Bim, thereby indirectly disturb sperm formation.
