实验性2型糖尿病心肌病大鼠模型建立的方法 被引量:15

Method to establish experimental rat model of type 2 diabetic cardiomyopathy

摘要 目的:采用持续高糖高脂膳食结合小剂量链脲佐菌素(STZ)多次腹腔注射,建立最佳类似人类2型糖尿病心肌病大鼠的造模方法。方法:把60只7~8周龄SD健康雄鼠随机分为3组:①对照组,20只,全程普通饲料喂养。②实验组1,20只,前8周高糖高脂饲料喂养,第2、4、6周注射STZ20mg/kg,后4周普通饲料喂养。③实验组2,20只,前8周与实验组1前8周处理相同,后4周继续高糖高脂饲料喂养。于第0周、第8周末、第12周末检测各组动物血糖、糖化血红蛋白、血脂变化,心功能改变情况。结果:第8周末,与对照组比较,实验组1、2血糖和血脂各指标明显增高(P〈0.01);左心室舒张末期内径(LVDD)变小、左心室后壁厚度(LVPw)和室间隔厚度(IVSD)增大(P〈0.05)。第12周末,实验组2与对照组、实验组1比较,血糖和血脂各指标明显升高(P〈0.01);LVDD值明显变小、左室短轴缩短率(LVFs)和射血分数(EF)降低、而LVPW和IVSD值明显增大(P〈0.01)。结论:采用持续高糖高脂膳食结合小剂量STZ多次腹腔注射,可成功建立2型糖尿病心肌病大鼠模型。 Objective..The sustained high sucrose and high fat diet combined with low dose of streptozotocin (STZ) intraperitoneal injection molding method of many times has the best similar to human type 2 diabetic cardiomyopathy in rats. Method: The 60 healthy male SD rats with 7--8 weeks old were randomly divided into three groups :(1) the control group ( n = 20 ) , fed ordinarily. (2) the experimental group 1 ( n = 20 ) , high sugar and high fat diet for 8 weeks , injection of 2 0 mg / kgSTZ at second , 4 th, 6 th week , then normal diet for last 4 weeks. (3) the experimental group 2 (n=20), the treatment for first 8 weeks was same to experimental group 1, high fat and high sugar diet for last 4 weeks. Blood glucose, glycosylated hemoglobin, blood lipids and change of heart function were detected at 0, 8, 12th weekend. Result:At the end of the eighth week, compared with the experimental group 1, 2, the index of glucose and lipid values were significantly higher in the control group (P(0.01), left ventricular end diastolic diameter (LVDD) decreased, left ventrieular posterior wall thickness (LVPW) and interventricular septal thickness (IVSD) increased (P〈0.05). At the end of the twelfth week, compared with control group and experimental group 1, the index of blood glucose and lipid values in the experimental group 2 were significantly higher (P〈0.01), LVDD values were significantly smaller, left ventricular fractional shortening (LVFS) and ejection fraction (EF) decreased, and LVPW and IVSD value significantly increased (P(0.01). Conclusion: The sustained high fat and high sugar diet combine with low dose STZ repeated intraperitoneal injection can be successfully established models of type 2 diabetic cardiomyopathy rats.
