高效毛细管电泳测定药品和人体血清中的氢溴酸加兰他敏

摘要 利用毛细管区带电泳法测定药品和人体血清中的氢溴酸加兰他敏。最佳实验条件是:50mmol/LNaH2PO4-H3P04,pH=2.5为背景缓冲溶液,分离电压为15kV,温度为25℃,检测波长为289nm。实验结果表明:氢溴酸加兰他敏在0.015-0.160mg/mL范围内呈良好的线性关系,r=0.9989,最低检出限是2.57×10-5mg/mL。日内和日间迁移时间和峰面积的相对标准偏差RSD分别是0.10%~0.22%、0.13%~1.10%和0.14%~0.34%、0.76%~1.63%,精密度和重现性均良好。该方法可简单、快捷地检测药品和人体血清中的氢溴酸加兰他敏。 The complex between Pd2. and 9-Bromophenanthrene (9-BrP) which offers remarkable enhancement to the fluores- cence of 9-BrP was found for the first time. The maximum excitation/emission wavelength of 9-BrP-Pd2+ are 247 and 364 nm, sepa- rately. According to the fluorescent enhancement theory, the coordination number of 9-BrP-Pd2+ was calculated to be 3, complex con- stant KA is 2.812× 10 4, the linear correlation coefficient is 0.9983. Pd2+, when the concentration is in the range of 1 × 10-7 to 2× 10-5mol. L 1, has a well Linear correlation with 9-BrP (R2=0.9962). The complex of Pd2+ with 9-BrP shows good selectivity and specifity, while the results of detecting Pd2+ in tap water sample and the standard addition test are also ideal, indicating the feasibility of using 9-BrP as a fluorescent probe for determination of Pd2+.
