摘要
目的研究壮筋续骨汤对大鼠股骨骨折后血清碱性成纤维细胞因子(bFGF)和转化生长因子(TGFβ1)水平以及骨折愈合的影响。方法成年健康雄性Wistar大鼠72只,应用电锯在股骨中段切断股骨制备骨折动物模型,壮筋续骨汤水煎液灌胃干预治疗。大体解剖观察骨折局部的愈合情况,组织病理学观察骨痂结构,酶联免疫吸附试验检测血清bFGF和TGFβ1水平。结果解剖观察发现,骨折后7d骨折断端主要为肉芽组织,14d为纤维性骨痂,21d可见骨性骨痂,模型组与治疗组未见显著差异,28d治疗组骨痂重塑质量明显优于模型组。大鼠血清bFGF和TGFβ1水平,模型组与治疗组在骨折后7、14、21d无显著差异,在28d治疗组明显高于模型组。结论壮筋续骨汤可能在骨折愈合后期通过维持血清bFGF和TGFβ1的水平、增强其活性、减缓其降解而促进骨折愈合。
Objective To investigate the effect of Zhuang Jin Xu Gu decoction (ZJXG decoction) on serum levels of basic fibroblast growth factor (bFGF) and transforming growth factorβ1(TGFβ1) and to explore its promoting healing of femoral fracture in rats. Methods Femur fractures were generated in 72 male adult Wistar rats by cutting femur transversely at middle point. ZJXG decoction was administered orally after surgery for 7-28d. The healing process was analyzed by gross anatomy and hematoxtlin-eosin (HE)staining. The serum levels of bFGF and TGFβ1 were detected by enzyme-linked immunobsorbent assay. Results Gross anatomy indicated that the fibrous callus tissue at the femoral fracture-end increased at 7-28 d following treatment with ZJXG Decoction, compared to control group. HE staining showed that the fibrous-granular tissue at the fracture-end changed gradually to fibrous, cartilaginous and osseous callus tissues. ELISA results showed that serum levels of bFGF and TGFβ1 increased significantly 28 days following treatment with ZJXG Decoction,compared to control group. Conclusion It is concluded that ZJXG Decoction could enhance the fracture healing by keeping the serum levels of bFGF and TGFβ1 and enhancing their activities and reducing their decomposition.
