摘要
目的建立高效液相色谱法测定大鼠血浆中的五味子醇甲,考察人参五味子颗粒与五味子稠膏中的五味子醇甲在大鼠体内的药动学差别。方法大鼠分别灌胃给予相当于五味子醇甲49.6mg/kg的五味子稠膏和人参五味子颗粒,血样用HPLC分析。采用C18色谱柱,甲醇-水(67:33)为流动相,检测波长250nm。结果五味子醇甲血浆浓度在0.22~27.00μg/mL之间线性关系良好,低、中、高浓度样品回收率分别为64.3%,70.2%和68.7%。大鼠灌胃五味子稠膏和人参五味子颗粒后的主要药动学参数分别为AUCo-t(26.67)和(30.77)mg/(h·L),AUCo-∞(29.44)和(41.64)mg/(h·L),tmax(0.86)和(0.83),Cmax(11.19)和(12.94),药一时曲线均符合二房室模型。结论五味子醇甲在大鼠体内药动学的研对中药复方制剂的药物动力学研究探索有一定进展。
Objective To investigate the pharmacokinetic differences of schisandrin in Ginseng schisandra granules and schisandra thick paste inside rats via high efficiency liquid chromatography used to test schisandrin in rat's plasma. Methods Rats were intragastrically administered schisandra thick paste and Ginseng schisandra granules,both of which contains 49.6 mg/kg of schisandrin.Their blood samples were analyzed by HPLC.C,s chromatographic column was applied.Methanol-water (67:33) was in mobile phase,and the testing wavelength was 250 nm. Results Linearity remained good when the blood concentration of schisandrin was between 0.22-27.00 μg/mL.The recovery rates of samples of low, medium and high concentrations were 64.3%,70.2% and 68.7% respectively.Main pharmacokinetic parameters after intragastric administration of schisandra thick paste and Ginseng schisandra granules for rats were AUCo-t (26.67) and (30.77) mg/ ( h · L ),AUC0- ∞ (29.44) and (41.64) mg/ ( h · L ),tmax (0.86) and(0.83),Cmax (11.19) and (12.94) respectively.The concentration-time curves all accorded to two-compartmental model. Conclusion The study on pharmacokinetics of schisandrin inside rats promotes and enlightens researches and explorations of pharmacokinetics of compound preparation of Chinese herbal medicine.
