金水鲜胶囊联合放射治疗对Lewis肺癌小鼠肿瘤生长和血管内皮生长因子表达影响的研究 被引量:2

Effect of Jinshuixian capsule in combination with radiotherapy on the tumor growth and expression of VEGF in Lewis lung cancer xenograft model

摘要 目的观察金水鲜胶囊联合放射治疗(简称放疗)对Lewis肺癌小鼠肿瘤生长及血管内皮生长因子(VEGF)表达的影响。方法建立Lewis肺癌移植瘤小鼠模型,随机分为6组,即空白对照组、金水鲜组、放疗组、先金水鲜后放疗组、先放疗后金水鲜组和同时金水鲜+放疗组,每组6只。放疗剂量为2Gy,金水鲜制成悬液后灌胃给药。游标卡尺测量瘤体长短径变化,绘制肿瘤生长曲线;采用实时聚合酶链反应(real-timePCR)和免疫组织化学法检测瘤体VEGF的表达。结果肿瘤体积在接受处理后明显减小。与空白对照组相比,先金水鲜后放疗组、先放疗后金水鲜组和同时金水鲜+放疗组肿瘤体积减小最为明显(分别为303.209 mm3±14.445 mm3,284.758 mm3±21.891 mm3,303.228 mm3±2.335 mm3),差异有统计学意义(P<0.05)。先金水鲜后放疗组、先放疗后金水鲜组和同时金水鲜+放疗组与空白对照组或放疗组比较,VEGF表达阳性率降低(分别为12.97%±0.82%,15.13%±1.31%,13.62%±0.83%),差异有统计学意义(P<0.05)。结论金水鲜胶囊与放疗联用对Lewis肺癌小鼠有抑制肿瘤的作用,其机制可能是下调VEGF的表达。 Objective To investigate the effect of Jinshuixian capsule combined with radiotherapy on the tumor growth and expression of vascular endothelial growth factor(VEGF) in mouse model with Lewis lung cancer. Methods Lewis lung cancer xenograft model was established and the tumor bearing mice were divided into 6 groups randomly: blank control group, Jinshuixian group, radiotherapy group, radiotherapy after Jinshuixian group, Jinshuixian after radiotherapy group, and Jinshuixian combined with radiotherapy group(n = 6). The radiation dose was 2 Gy and Jinshuixian suspension was intra-gastrically administrated into mice. Tumor line of apsides was measured by venire calipers and tumor volume was calculated. Immunohistochemisty and real-time polymerase chain reaction were adopted to check the expression of VEGF. Results Tumor volume was significantly reduced after treatment. The maximum reduction of tumor volume was (303.209 ± 14.445) mm^3 in radiotherapy after Jinshuixian group, (284.758 ± 21.891) mm^3 in Jinshuixian after radiotherapy group, and (303.228 ± 2.335) mm3 in Jinshuixian combined with radiotherapy concomitantly group, which were significantly decreased as compared with blank control group (P 〈 0.05). Compared with blank control group and radiotherapy group, the positive expression rates of VEGF in radiotherapy after Jinshuixian group(12.97 % ± 0.82 %), Jinshuixian after radiotherapy(15.13 % ± 1.31%), and Jinshuixian combined with radiotherapy concomitantly group(13.62 % ± 0.83 %) were significantly lower(P 〈 0.05). Conclusion It is demonstrated that Jinshuixian capsule in combination with radiotherapy could inhibit the growth of Lewis lung cancer, the inhibition of VEGF expression may be one of the mechanisms.
