摘要
目的:探讨补糖对一次较大强度运动后血清Toll样受体4(TLR4)水平和白细胞表面TLR4mRNA表达的影响。方法:随机选取8名健康男性青年进行两次跑台运动,实验时间为两周,两次测试间隔1周。运动前和运动中随机补充下列饮料的一种:含糖饮料(补糖组)或对照饮料(对照组),含糖饮料的补充量为0.8 g/kg体重。摄取一种饮料后,在跑台上以75%VO2max的强度跑步60分钟。分别在安静、运动后即刻、运动后1小时、运动后2小时和运动后24小时测定血清IL-6、TLR4和NF-κB水平及白细胞TLR4 mRNA相对表达量。结果:(1)运动后,对照组白细胞计数显著升高,运动后2小时非常显著高于安静水平,运动后24小时回落到安静值;(2)运动后即刻,对照组血清IL-6水平有升高趋势,补糖组变化不明显;(3)运动后即刻,对照组血清TLR4水平显著下降,补糖组下降不明显,补糖组显著高于对照组;运动后2小时,对照组和补糖组白细胞TLR4 mRNA相对表达量显著低于安静值,在运动后1小时补糖组显著高于对照组。(4)运动后即刻,对照组血清NF-κB水平比安静值升高了93%,补糖组升高幅度为43%,低于对照组。结论:一次较大强度运动能引起血清TLR-4水平和白细胞TLR-4mRNA表达的下降,补糖能缓解运动造成的血清TLR-4水平降低和白细胞TLR-4 mRNA表达降低。
Objective To observe the effect of carbohydrate supplementation on the TLR4 level in serum and TLR4 mRNA expression in ]eukocytes of male college students during and after heavy exercise. Methods 8 healthy untrained male subjects were recruited in the trial. Each subject randomly consumed one of the following beverages: carbohydrate containing beverage (CHO group)or placebo (PLA group). The carbohydrate containing beverage was taken by each subject with the dosage of 0.Sg carbohydrate/kg body mass. Each subject underwent two trials with an interval of one week. The trial consisted of a running at 75%VOEmax for 60 minutes on a treadmill. Blood sample was taken before exercise,immediately after exercise,and 1 hour,2 hours and 24 hours after exercise for analyzing the contents of IL-6,TLR-4 and NF-KB in serum and the mRNA expression of TLR4 in jeukocvtes. Results(1)WBC count increased significantly immediately after exercise,especially 2 hours after exercise,and then declined until 24 hours after exercise.(2)The level of IL-6 in PLA group increased insignificantly, and the blood glucose remained unchanged in ClIO group. (3)The level of TLR4 in serum decreased markedly in PLA group immediately after exercise,and that in CHO group was higher than PLA group. The mRNA expression in leukocytes decreased 2 hours after exercise in both CHO and PLA groups,but was higher in CHO group than in PLA group 1 hour after exercise.(4)Compared with the base line,the level of NF-KB increased by 93% in PLA group, and by 43% in CHO group. Conclusion The level of TLR4 in serum and mRNA expression of TLR4 in leukocytes decreased after 1-hour treadmill exercise, which could be attenuated by carbohydrate supplementation.
