摘要
【目的】探讨油茶芽苗砧嫁接愈合过程中砧穗相关生理指标的动态变化规律,为油茶芽苗砧嫁接亲和性研究提供理论依据。【方法】以油茶优良品种‘XL1’芽苗砧嫁接苗为材料,研究芽苗砧嫁接愈合过程中(0~80d)砧木与接穗中可溶性蛋白、可溶性糖含量及过氧化物酶(POD)活性的变化。【结果】砧木可溶性蛋白含量呈现"降-升-降"的变化趋势,为嫁接体接口愈合提供能量,接穗可溶性蛋白含量在31.94~40.39mg/g,变化幅度不大;在输导组织分化前,接穗可溶性糖含量先降低后升高,砧木可溶性糖含量保持平稳,且始终低于接穗;在形成贯通接穗和砧木的维管束桥时,砧木的POD活性较高,接穗的POD活性始终很低,维持在0.11~0.62U/mg。【结论】在油茶芽苗砧嫁接愈合过程中,砧木的可溶性蛋白含量与POD活性高于接穗,而接穗的可溶性糖含量高于砧木。
[Objective] This study focused on the dynamic changes in physiological indexes of Camellia oleifera nurse seed graft in the grafting and recovering process. [Method] Dynamic changes of physiologi- cal indexes including soluble protein,soluble sugar,and POD activity of C. oleifera (XL1) scion and stock in the grafting and recovering process (0--80 d) were studied. The soluble protein content was detected by coomassie brilliant blue method, the soluble sugar content was detected by anthrone colorimetry, and the POD activity was detected by guaiacol method. [Result] The soluble protein content of stock presented a trend of "decreasing-increasing-decreasing", indicating that it provided nutrients for the grafts. The soluble protein content of scion changed slightly within range of 31.94--40.39 mg/g. Before the differentiation of conducting tissue, the soluble sugar content of scion increased after initial decrease while that of stock re- mained unchanged and was always lower than that in scion. When the vascular tissue was formed, POD ac- tivity of stock was relatively high,while POD activity of scion was at a low level of 0. 11--0. 62 U/mg. [Conclusion] In the grafting and recovering process, soluble protein content and POD activity of stock were higher than that of scion,while the soluble sugar content of scion was higher than that of stock.
