摇晃蛋白(Reelin)的剪切蛋白预测

摘要 【目的】探索生物信息学方法在Reelin剪切蛋白查找中的应用,并筛选大脑皮质中可能对Reelin有剪切作用的蛋白。【方法】以ADAMTS-4蛋白为基准,利用生物信息学方法,以功能域查找相似功能蛋白,并结合同源性比对方法从中筛选出剪切Reelin的候选蛋白,并对候选蛋白的理化性质、亚细胞定位、功能域、二级结构和组织定位进行分析。【结果】ADAMTS-2、ADAMTS-3、ADAMTS-10、ADAMTS-16与ADAMTS-4有相似的理化性质,且均属于分泌蛋白,在大脑皮质中可能有分布。ADAMTS-2、ADAMTS-3、ADAMTS-10、ADAMTS-16包含ADAMTS-4的功能域,且功能域内的二级结构相似。【结论】筛选到了可能对Reelin有剪切作用的蛋白ADAMTS-2、ADAMTS-3、ADAMTS-10和ADAMTS-16。 [Objective] This study attempted to use bioinformatics tools to predict proteases of Ree in cerebral cortex. [Method] Using ADAMTS-4 protein as the benchmark, candidate proteases for Ree were screened by bioinformatics method, based on search of similar functional proteins and comparison in in of homologous proteins. The chemicophysical properties, secondary structure, functional domains, cellular lo- calization and tissue distribution of candidate proteases were analyzed as well. [Result] ADAMTS-2, AD- AMTS-3, ADAMTS-10,ADAMTS-16 and ADAMTS-4 were secretory proteins with similar physicochemi- cal properties and they might distribute in the cerebral cortex. ADAMTS-2, ADAMTS-3, ADAMTS-10, ADAMTS-16 contained same functional domains as ADAMTS-4 and their secondary structures were simi- lar as well. [Conclusion] ADAMTS-2, ADAMTS-3, ADAMTS-10, ADAMTS-16 were selected proteases for Reelin.
