响应面法优化雷公藤中雷公藤甲素的超声提取工艺 被引量:2

Optimization of Ultrasonic Extraction Process for Triptolide from Tripterygium wilfordii by Response Surface Methodology

摘要 目的:优选雷公藤中雷公藤甲素的超声提取工艺。方法:以雷公藤甲素得率为指标,采用响应面设计考察料液比、乙醇体积分数、超声时间及超声温度对雷公藤甲素提取工艺的影响。采用HPLC测定雷公藤甲素含量,流动相乙腈-水(33∶67),检测波长218 nm。结果:最佳提取工艺为料液比1∶12,乙醇体积分数70%,提取温度35℃,提取时间30 min;雷公藤甲素得率达0.037 9 mg·g-1,与模型预测值(0.038 6 mg·g-1)较为接近。结论:优选的超声提取工艺稳定可靠,与常规回流提取法相比优势明显。 Objective: To optimize ultrasonic extraction process of triptolide from Tripterygium wilfordii. Method: With yield of triptolide as index,response surface methodology was adopted to optimize extraction technology by taking material-liquid ratio,ethanol concentration,extraction time and ultrasonic temperature as factors.HPLC was adopted to determine the content of triptolide with mobile phase of acetonitrile-water(33:67) and detection wavelength at 218 nm. Result: Optimal extraction conditions were as follows:solid-liquid ratio of 1:12,ethanol concentration 70%,extraction temperature at 35 ℃,extraction time 30 min;under these conditions,yield of triptolide was 0.037 9 mg·g^-1,which was close to the predicted value(0.038 6 mg·g^-1). Conclusion: This optimized extraction process was stable and reliable,which had obvious advantages by comparing with conventional reflux extraction method.
