摘要
目的:建立赤芍、大黄药对不同配伍比例的高效液相指纹图谱。方法:采用Agilent Eclipse XDB-C18(4.6 mm×250 mm,5 μm)色谱柱,以乙腈(A)-0.1%磷酸水(B)为流动相进行梯度洗脱,流速1.0 mL·min^-1,检测波长267 nm(0-13 min),258 nm(13-20 min),230 nm(20-45 min),254 nm(45-100 min),柱温30 ℃。结果:建立了赤芍、大黄药对不同配伍比例的高效液相指纹图谱。以芍药苷为参照峰,标定18个共有峰,指认了其中9个色谱峰。通过赤芍、大黄5个比例指纹图谱的研究,初步确定了两药材主要成分的归属,以芍药苷与大黄酸峰面积比值为指标确定其配伍比例。结论:该方法准确可靠、重复性好,可用于为赤芍、大黄药对提取物的成分鉴别及配伍和质量控制提供依据。
Objective: To develop an HPLC fingerprint for the different compatibility proportion in Radix Paeoniae Rubra and Rhei Radix et Rhizoma. Method: Agilent Eclipse XDB-C18 (4.6 mm×250 mm, 5 μm) was adopted;the mobile phase was acetonitrile (A)-0.1% phosphoric acid solution (B) with gradient elution at a flow rate of 1.0 mL·min-1,and the detection wavelength was set at 267 nm (0-13 min), 258 nm (13-20 min), 230 nm (20-45 min), 254 nm (45-100 min). Result: An HPLC fingerprint for the different compatibility proportion in Radix Paeoniae Rubra and Rhei Radix et Rhizoma was developed. A total of 18 chromatographic peaks and 9 chromatographic peaks were identified. Through studying 5 proportion of fingerprint of Radix Paeoniae Rubra and Rhei Radix et Rhizoma, the main components of two herbs belonging were preliminary identified. Conclusion: This method is dependable, simple and practical, and it may be used to identify component and control quality of Radix Paeoniae Rubra and Rhei Radix et Rhizoma.
