六神曲中真菌的分离与鉴定 被引量:22

摘要 目的:对传统工艺发酵六神曲中真菌进行分离与鉴定研究。方法:采用平板划线法和菌丝顶端纯化法从传统发酵六神曲分离真菌,使用生物显微镜进行显微形态学鉴定,DNA测序进行分子生物学鉴定。结果:从传统发酵六神曲分离并确定3株真菌分别为曲霉属真菌黄曲霉菌,青霉属真菌产黄青霉菌以及枝孢属真菌枝孢霉菌。黄曲霉菌和产黄青霉菌发酵六神曲气味纯香,表面遍布白衣略有黄衣出现与传统发酵相似;枝孢霉菌发酵六神曲产生霉腐气味,曲块表面暗灰褐色与传统发酵差别大,推测枝孢霉菌为六神曲发酵中产生的杂菌。结论:从传统方法发酵的六神曲分离得到3株真菌,经形态学鉴定及分子生物学鉴定,确定3株真菌分别为曲霉属真菌黄曲霉菌,青霉属真菌产黄青霉菌以及枝孢属真菌枝孢霉菌,为六神曲有效菌群的筛选提供了重要的物质基础研究。 Objective: To study the isolation and identification of Fungi in Medicated Leaven. Method: Using streak plate method and the hyphal tip purification methods to isolate fungi from Medicated Leaven, and using biological microscope microscopic method to identify morphology study, along with DNA sequencing for molecular identification. Result: There strains were identified three strains of fungi were Aspergillus flavus, Penicillium chrysogenum and Cladosporium fungus by morphology and molecular biology identification. The Aspergillus and P. chrysogenum fermentation of medicated leaven, it smells and trait similarity and traditional fermentation. C. fungus fermentation of medicated leaven produce rotting smell, its characters is also great different from traditional fermented difference. Conclusion: The results provide material basement for further studies related to Medicated Leaven.
