桂枝茯苓胶囊治疗原发性痛经的实验研究 被引量:14

摘要 目的:研究桂枝茯苓胶囊对痛经小鼠及大鼠的治疗作用。方法:复制前列腺素F2α(PGF2α)致小鼠痛经模型,观察桂枝茯苓胶囊对小鼠扭体次数、血浆β-内啡肽(β-EP)、6-酮-前列腺素F1α(6-Keto-PGF1α)含量的影响;复制缩宫素(OT)致大鼠痛经模型,观察桂枝茯苓胶囊对大鼠扭体次数、子宫组织前列腺素F2α(PGF2α)、前列腺素E2(PGE2)的影响。结果:与模型组比较,桂枝茯苓胶囊高、中剂量组可显著减少前列腺素F2α引起的小鼠扭体次数(P<0.05),显著升高血清中β-EP含量(P<0.05),桂枝茯苓胶囊高剂量组能显著升高血清中6-Keto-PGF1α含量(P<0.05);与模型组比较,桂枝茯苓胶囊高剂量组可显著减少缩宫素引起的大鼠扭体次数,显著降低大鼠子宫组织PGF2α含量,桂枝茯苓胶囊高、中剂量组PGF2α/PGE2比值降低。结论:桂枝茯苓胶囊治疗痛经作用机制可能与调节体内β-EP、6-Keto-PGF1α、PGF2α、PGE2水平有关。 Objective: To investigate the effect and mechanism of Guizhi Fuling capsule on primary dysmenorrheal (PD) in mice. Methods: The effects of different dosage of Guizhi Fuling capsule on writhing frequency, the serum level of β-endorphin (β-EP), 6-Keto-PGF1α were observed in PGF2α-induced female mice. The effects of different dosage of Guizhi Fuling cap- sule on writhing frequency, the level of prostaglandin F2α(PGF2α), prostaglandinE2(PGE2)in the uterus were observed in oxy- tocin-induced female rats; Results: Compared with the model group, the writhing frequency in mice significantly decreased in higher and middle dosage groups of Guizhi Fuling capsule (P〈0.05), and the serum level of β-EP,6-Keto-PGF1α were signifi- cantly increased in mice (P〈0.05). As compared with the model group, the writhing frequency in rats significantly decreased in higher and middle dosage groups of Guizhi Fuling capsule (P〈0.05), the level of PGF2α and the ratio of PGF2α/ PGE2 in rat uterus was significantly decreased (P〈0.05). Conclusion Guizhi Fuling capsule has an analgesic effect on dysmenorrhea in- duced by prostaglandin F2α in mice and oxytocin in rats,and its mechanism may be regulate with the level of β-EP,6- Keto-PGF1α, PGF2α, PGE2.
