兔脂肪间充质干细胞向胰岛素样分泌细胞分化及鉴定

摘要 脂肪间充质干细胞(adipose-derived mesenchymal stem cells,ADSCs)最早由Zuk[1]等人于2001年从抽脂术中抽取的脂肪组织中分离培养,近年来已成为医学领域乃至整个生命科学领域的研究热点.ADSCs是一种来源于脂肪组织中的多能干细胞,和骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)类似,也有很强的增殖能力和分化潜能.ADSCs能在不同的诱导微环境下向不同的组织细胞(如脂肪细胞、成骨细胞、内皮细胞、心肌细胞和神经细胞等)分化[1].本实验在体外分离培养兔的脂肪间充质干细胞,通过相应的试剂使其诱导分化为类胰岛素分泌细胞,并研究其生物学特性,以期为临床采用ADSCs治疗Ⅰ型糖尿病提供理论依据.
