摘要
目的建立溶藻弧菌腹腔注射感染小鼠模型并对动物分离株进行药物敏感性试验分析。方法采用半数致死量试验确定最适感染菌量和感染时间,并通过尾血细菌培养和微生物自动鉴定仪确定溶藻弧菌感染,同时对分离菌株进行抗菌药物敏感性试验。结果小鼠感染溶藻弧菌3h后开始出现症状,且症状的轻重与注射的菌量成正相关。溶藻弧菌E0666株感染昆明小鼠的半数致死量为109cfu,平均存活时间是20h。从感染组小鼠尾血中分离的细菌对临床常用的大多数抗生素敏感,但对氨苄西林和头孢噻吩耐药。结论溶藻弧菌动物感染模型的建立及动物分离株药敏试验研究为探讨溶藻弧菌的致病机制及其治疗和防治都具有重要意义。
Objective To establish a Vibrio alginolyticus (V.alginolyticus) infected mouse model by intraperitone al injection and analysis of the antibiotic sensitivity testing with animal isolates.Methods The median lethal dose(LD50) test was performed to determine the optimal microbial infection and infection time,and the bacteria culture of tail blood and microbial identification instrument to determine V.alginolyticus infection.Then antibiotic sensitivity testing was done for the isolated strain.Results After V.alginolyticus infection for 3 h,the mice suffered different dam age symptoms,and the degree of injury was closely relative to the amount of bacteria.The LD50 of V.alginolyticus E0666 strain was 109 cfu in kunming mice,and the average survival time was 20 h.Bacteria isolated from tail blood of mice were sensitive to most of the clinical commonly used antibiotics,but drug resistant to ampicillin and cefalotin.Conclusion The study of establishing V.alginolyticus infected mouse model and antibiotic sensitivity testing played an important role in researching the pathogenic mechanism of V.alginolyticus and its treatment and prevention.
