摘要 设计了3种从土壤中直接提取微生物DNA的方法,并通过PCR扩增和DGGE电泳比较了不同方法所提取DNA的质量以及对后期分子生物学研究的适用性。结果表明,化学法和酶法相结合的提取方法所获得的DNA完整性最好,适用于PCR扩增,并且通过DGGE电泳分离出最丰富的条带,是一种高效的提取土壤微生物DNA的方法。 Three methods were designed to extract microbial DNA direcdy from the soil, and the different extraction methods were evaluated comprehensively for the DNA quality and applicability of the molecular biology by PCR and DGGE. The re suits suggested that the combination of enzymatic and chemical methods could obtain the highest DNA yield and integrity, which was suitable for PCR, and the most abundant bands could be obtained by DGGE. This development of the highly ef fective DNA extraction method provided a foundation for studying in molecular ecology of soil microbes.
