摘要
甲羟戊酸激酶(mevalonate kinase,MK)是萜类化合物生物合成甲羟戊酸(mevalonate pathway,MVA)途径的限速酶之一。为了探明EuMK对杜仲胶生物合成的调控机制,对EuMK所编码蛋白质氨基酸序列的理化性质、跨膜结构域、疏水性/亲水性、蛋白质二、三级结构、结构保守域以及基因组内含子和外显子结构特征等进行了全面的生物信息学分析,结果发现EuMK蛋白质序列均存在3个与甲羟戊酸底物结合及催化反应有关的保守结构域;EuMK的氨基酸组成中性的疏水性蛋白居多数,属于稳定的疏水性蛋白;无明显的跨膜结构,均在膜外;二级结构为混合型结构的蛋白质。EuMK均含有4个外显子,3个内含子;EuMK基因家族启动子顺式作用元件潜在功能的预测分析发现,含有大量基本顺式作用元件TATAbox,CAATbox,还含有光(AE-box、CGT-motif、Box 4、G-box)、生长素(ERE、ABRE、TCA-element)响应等多个顺式调控元件。
Mevalonate kinase (MK) is one of the rate-limiting enzymes in mevalonate pathway (MVA) of terpenoid biosynthetic. In order to ascertain regulatory mechanism of EuMK for biosynthetic mechanism of Eucommia rubber, EuMK was analyzed and predicted by the method of bioinformatics from the aspects of physiochemical property, transmembrane domain, hydrophobicity or hydrophilicity, and so on. The results show that EuMK have three domains related to mevalonate substrate binding and catalytic reactions. Major amino acids in EuMK are of the neutral hydrophobicity proteins, so EuMK is a stable hydrophobicity protein, and it has no clear transmembrane domain. Secondary structure of EuMK protein belong to hybrid structure. EuMK have four exons, three introns. Some potential functions of the promoter cis-acting elements in EuMK gene family were forecasted. EuMK gene family contains a lot of basic cis-acting elements and a number of cis-regulatory elements, such as TATAbox, CAATbox, light response (AE-box, CGT-motif, Box 4, G-box), auxin response (ERE, ABRE, TCA-element), and so on.
