摘要
杜仲肉桂酸-4-羟化酶(C4H)是植物苯丙素类物质合成的关键酶之一。为了探明生物合成杜仲绿原酸的分子调控机制,对杜仲C4H基因全长cDNA进行了系统的生物信息学研究,鉴定了2个基因家族成员,分别为EuC4H1和EuC4H2,其全长cDNA分别为1 641 bp和1 611 bp;编码547和537个氨基酸,相对分子质量分别为62.94 kD和60.96 kD,等电点分别为9.34和8.85,均为疏水性不稳定蛋白质,其中EuC4H2是分泌蛋白;二级结构均是以α-螺旋、β-折叠、螺环结构并存的混合型结构,都属于Cypx超家族。
Cinnamic acid-4-hydroxylase (C4H) is one of a key enzyme in the synthesis ofphenylpropanoid substances. In order to study molecular regulator mechanism of chlorogenic acid biosynthesis in Eucommia ulmoides, full-length cDNA of EuC4H was analyzed by the tool of bioinformatics. The EuC4H gene family was identified, which comprises two members of EuC4H1 and EuC4H2. Full-length cDNA ofEuC4Hland EuC4H2 are 1 641 bp and 1 611 bp respectively. The proteins encoded by EuC4H1 and EuC4H2 are 547 aa and 537 aa, relatively molecular mass are 62.94 kD and 60.96 kD, and theoretical isoelectric point are 9.34 and 8.85 respectively. Both of them are instable hydrophobic proteins, and EuC4H2 is a secreted proteins. The secondary structures of the proteins are of the hybrid structure of α-helix, β-sheet and random coil, belonging Cypx superfamily.
