摘要 苯丙素类化合物的合成过程中,苯丙氨酸解氨酶(PAL)是植物苯丙烷类合成的第1个关键酶。为了解杜仲苯丙氨酸解氨酶基因序列信息,为杜仲苯丙素类高效合成的分子调控提供科学依据,对杜仲PAL基因cDNA和基因组全长序列特征进行了系统的生物信息学分析。结果表明,杜仲的EuPAL1基因的全长cDNA,其长度为2 133 bp;该基因共编码711个aa,酶蛋白相对分子质量77.14 kD,等电点6.24,属于稳定疏水性蛋白;编码蛋白二级结构是以α-螺旋和螺环结构为主的混合型结构,属于lyase-I-like蛋白超家族。 Phenylalanine ammonia lyase (PAL) is the first key enzyme during synthesis of phenylpropanoid compounds. In order to understand gene sequence information and further research the gene function of phenylalanine ammonia lyase in Eucornmia ulrnoides, and to provide a scientific basis for molecular regulation of efficient synthetic of phenylpropanoids, cDNA and full length genome sequence of EuPAL were systematically analyzed by the tool of bioinformatics. The results show that full-length cDNA of EuPAL1 is 2 133 bp and the gene encode 711 amino acids. The relative molecular mass of the EuPALI protein is 77.14 kD, its theoretical isoelectric point is 6.24, and the protein belongs to the stable hydrophobic protein. The secondary structure of the protein mainly is α-helix and random coil, belonging lyase-I-like superfamily.
