白介素-6、基质金属蛋白酶-2在早期胚胎发育中的作用 被引量:10

摘要 目的:探讨白介素-6(IL-6)及基质金属蛋白酶-2(MMP-2)在胚胎发育中的可能作用。方法:对30例不明原因胚胎停止发育病例(研究组)和30例正常早孕吸宫流产病例(对照组)的绒毛组织及血清进行研究。采用实时荧光定量逆转录聚合酶链式反应技术(Real time-PCR)和免疫组织化学技术(SP法)检测两组绒毛组织中IL-6和MMP-2 mRNA的表达和蛋白定位、表达强度;采用双抗体酶联免疫夹心法(ELISA法)测定IL-6在两组血清中的浓度。结果:绒毛组织中,IL-6与MMP-2在合体及绒毛滋养层细胞中均有表达,Real time-PCR及免疫组化结果均显示IL-6在研究组中的表达水平低于对照组,MMP-2在研究组中的表达水平高于对照组,差异均有统计学意义(P<0.05);IL-6在血清中的表达水平组间无统计学差异(P>0.05)。结论:孕早期绒毛组织中IL-6和MMP-2的正常表达对维持早期胚胎发育起到一定作用。 Objective To explore the relationship between the Interleukin 6, matrix metalloproteinases 2 and early embryo arrest. Methods Real time-PCR was used to measure the mRNA expression of IL-6 and MMP2 and immunohistochemistry (IHC, SP method)was used to measure the location and expression of the two different kinds of protein in villus. ELISA was used to measure the level of IL-6 in serum. Results Real-time PCR and IHC showed that the expression levels of IL-6 was significantly lower in experimental group than in control group, and MMP-2 was significantly higher than the control group (P 〈 0.05). Differenc of IL-6 level in serum between the two groups was not statistically significant (P 〉 0.05). Conclusion Proper expressions of IL-6 and MMP-2 in the villus tissue play a key role in the maintenance of early pregnancy.
