牦牛JHDM2A基因的克隆测序及其生物信息学分析 被引量:1

摘要 利用RT-PCR克隆技术对牦牛JHDM2A基因进行cDNA克隆测序,并用生物信息学软件分析该基因的编码区序列、蛋白结构及进化关系。结果表明:①牦牛JHDM2A基因cDNA序列大小为4 605bp,其中CDS区序列3 969bp,编码氨基酸1 323个。牦牛JHDM2A基因与人、小鼠、褐鼠、原鸡等物种该基因序列比对,一致性分别为90.3%、86.9%、86.3%、69.9%,在物种间具有较高的一致性。②牦牛的JHDM2A蛋白存在JMJC结构域,属于JMJD家族的一员,该蛋白是一种弱碱性、无跨膜结构的游离蛋白,不定位于细胞的任何部位,进化速度慢、保守性强。以上结果为进一步从分子水平上了解牦牛JHDM2A基因和JHDM2A蛋白的结构、功能提供了理论基础。 JHDA/I2A gene is one of the key factors which stem cell functional activity and embryo development. have epigenetic effects on spermatogenesis, In this study, the cDNA sequence of the JHDM2A gene in yak (Bos grunniens) was cloned by RT-PCR method, and the coding region se- quence, protein structure and phylogenetic relationship were analyzed by bioinformatics software. The results showed that the cDNA length ofJHDM2A gene was 4 605 bp and its CDs sequence was 3 969 bp encoding a putative protein of 1 323 amino acid (AA) residues. The nucleotide sequence identity of theJHDM2A gene between human, mouse, brown rat and chicken were 90.3%, 86.9%, 86. 3%, 69.9% respectively. The data suggested that the gene from the yak were highly conserved with other species at the nucleic acid level. JHDM2A protein in yak has a JMJC domain and belongs to JMJD family. It is a weak alkaline protein without transmembrane structure and sub-cellular localization peptide. The results provide theoretical bases for further study on yakJHDM2A gene andJHDM2A protein structure, function from molecular level.
