白芨MYB类转录因子PAP1基因的克隆与表达分析 被引量:6

摘要 花青素对植物的生长发育具有重要的作用。MYB家族转录因子参与花青素合成代谢的调控。利用同源基因克隆的方法,根据其他物种中基因的同源序列设计特异性引物,利用PCR技术从白芨基因组中克隆到MYB转录因子PAP1,cDNA全长844bp,经DNAStar软件分析,PAP1无内含子,包含一个完整的阅读框,编码184个氨基酸。不同组织的表达特征分析发现,BsPAP1在花中的表达较强,在块根状假鳞茎和嫩蒴果中的表达次之。蛋白质特征预测及蛋白序列结构分析发现PAP1具有MYB转录因子典型的DNA结合区域。序列同源比对表明该基因与芜菁、花椰菜等的PAP1基因有很高的相似性,推测BsPAP1是MYB家族的成员之一。 In plants, anthocyanins play an important physiological role in growth and development. Regulation of MYB family transcription factor involved in anthocyanin biosynthesis. In this studyBs- PAP1 was cloned from Bletilla striata based on homologous gene from other species. BsPAP1 was 844 bp in length without intron. RT-PCR analysis showed that BsPAP1 was expressed dominantly in flower followed by bulb and germinal capsule. Protein characteristics and sequence structure predicted that it was a protein with Myb-type HTH DNA-binding domain. BsPAP1 was identified as a PAP1 gene according to its high identity with Brassica rapa subsp, rapa cultivar Tsuda (accession. HQ337792)and Brassica oleracea var. botrytis cultivar Stovepipe(accession: GM219987). It was as- sumed that BsPAP1 was a member of MYB family.
