摘要
目的观察扶正解毒方对肺炎链球菌感染大鼠炎性细胞因子及免疫指标的影响,探讨扶正解毒法对肺感染后免疫失衡的调控机制。方法气管内注射肺炎链球菌建立肺炎感染模型,分别用扶正解毒中药、头孢呋辛酯、扶正解毒中药+头孢呋辛酯干预,分别于给药第4日和第7日取材。观察各组肺组织形态学,ELISA检测血清及肺组织中肿瘤坏死因子-α(TNF-α)、白介素-17(IL-17)的表达;West blot检测脾组织中CD4+、CD8+水平。结果与正常组相比,模型组肺组织与血清中TNF-α、IL-17含量增高(P<0.05)。各治疗组TNF-α、IL-17水平降低,CD4+、CD8+水平增高,免疫紊乱状态改善。结论扶正解毒中药可以降低炎性细胞因子水平、增强免疫功能,改善动物生存状态。扶正解毒中药对肺炎的治疗机制可能与降低炎性细胞因子水平,减轻炎性反应,增强免疫功能有关。
Objective:To observe the influence of Fuzhengiedu Decoction on inflammatory cytokines and immune function in streptococcus pnenmonia rats by.Methods:After pnenmonia infection models being establised by streptococcal pnenmonia tracheal injection,the experimental animal models were treated with Fuzhengjiedu Decoction,cefuroxime axetil and Fuzhengjiedu Decoction plus cefuroxime axetil.After 4 days and 7 days,concentrations of the TNF-α and the IL-17 were determined in the serum and lung tissue by ELISA.Meanwhile,the CD4+ and the CD8+ levels were detected in the spleen tissue by west blot.Results:Compared with the normal gorup,concentrations such as TNF-αand the IL-17 were increased significantly in the model group (P< 0.05).Treatment had a negative regulatory role on TNF-α and IL-17 (reduced) and a positive regulatory role on CD4+ and CD8+(increased) respectively,and could adjust the immune state of disorder.Conclusion:Fuzhengjiedu Decoction could reduce the levels of inflammatory cytokines,enhance the function of the immune and improve animal survival.The mechanism of treating pnenmonia may reduce the levels of inflammatory cytokines and enhance the immune function.
