摘要
目的观察金荞麦水提物对慢性阻塞性肺疾病(COPD)大鼠模型肺纤维化的影响。方法改良烟熏法复制COPD模型,分正常组、模型组、金荞麦水提物低剂量组、金荞麦水提物高剂量组4组,观察支气管和肺组织病理,免疫组织化学检测基质金属蛋白酶(MMP-9)、基质金属蛋白酶组织抑制因子(TIMP-1),放射免疫法血清上清液检测转化生长因子(TGF-β)、Ⅲ型前胶原肽(PⅢ)。结果与模型组比较,金荞麦水提物组能显著减轻大鼠支气管、肺组织的病理变化,以高剂量最为明显,抑制MMP-9和TIMP-1的过度表达,升高MMP-9/TIMP-1,降低血清TGF-β和PⅢ的水平。结论金荞麦水提物能改善COPD大鼠模型的肺纤维化。
Objective:To study the effect of the water extract of Fagopyrum dibotrys on COPD model of rats with pulmonary fibrosis.Methods:Rats were randomly divided into normal group,model group,and Fagopyrum dibotrys water-extraction low and high dose groups.Pathological morphological changes of lung and bronchus were observed.The expressions of MMP-9 and TIMP-1 were detected by immunohistochemistry method.The expressions of TGF-β and P Ⅲ were detected in serum supernatant by radioimmunoassay method.Results:Compared with normal group,Fagopyrum dibotrys water extract had significant improvement effect on the lung and bronchus pathomorphism changes of rats,especially in high dose group and inhibited excessive expressions of MMP-9/TIMP-1,increased the expression of MMP-9/TIMP-1,and decreased the expression of TGF-β and PⅢ in serum.Conclusion:Fagopyrum dibotrys water extract can improve the COPD model of rats with pulmonary fibrosis.
