摘要
目的探讨南京地区汉族人群中编码人类中性粒细胞抗原-3(human neutrophil antigen-3,HNA-3)的SLC44A2基因的多态性及等位基因频率分布。方法对南京地区107位随机献血者的SLC44A2基因采用聚合酶链反应(polymerase chain reaction,PCR)方法扩增其相关外显子及侧翼内含子,并进行测序分析。结果 HNA-3a/3a有46例,HNA-3a/3b 47例,HNA-3b/3b 14例,实验结果符合Hardy-Weinberg遗传定律。HNA-3a和HNA-3b在南京地区的频率分别为0.649 5和0.350 5。结论南京地区汉族人群HNA-3具有多态性,此多态性可能造成的抗-HNA-3a相关风险在临床上应引起重视。
Objective To investigate the polymorphism and frequency distribution of SLC44A2 alleles encoding HNA-3 antigen in Nanjing Han population. Methods 107 random blood donors' SLC44A2 relevant exons and adjacent introns were amplified by PCR method and sequencing analysis. Results 46 cases HNA-3a/3a,47 cases HNA-3a/3b and 14 ca- ses HNA-3b/3b were found and all statistics were consistent with the Hardy-Weinberg equilibrium law. The frequency dis- tribution of HNA-3a and HNA-3h was 0. 6495 and 0.3505 in Nanjing Han population, respectively. Conclusion The dis- tribution of HNA-3 is polymorphie in Nanjing Han population. The clinical risk of anti-HNA-3a should bring to the atten- tion.
