白头翁汤及其主要成分对脂多糖诱导的内皮细胞体外分泌IL-1α和IL-6及ET-1的影响 被引量:8

摘要 为探讨白头翁汤及其主要成分对细菌内毒素(LPS)诱发的病理效应的药理作用,培养内皮细胞至单层融合后加入内毒素,3 h后加入高(10 mg·mL-1)、中(5 mg·mL-1)、低(1 mg·mL-1)3种剂量的8种药物,继续培养21 h后收集细胞上清液,用ELISA方法测定IL-1α、IL-6和ET-1的含量。结果表明:白头翁汤及其成分白头翁皂苷B4、小檗碱、巴马汀可以极显著或显著抑制LPS诱导的IL-1α高水平分泌,白头翁汤及其成分秦皮甲素、秦皮乙素、白头翁素可以极显著或显著抑制LPS诱导的IL-6高水平分泌,白头翁汤及其成分秦皮乙素、白头翁素可以极显著或显著抑制LPS诱导的ET-1高水平分泌。结论:白头翁汤及其主要成分具有潜在的抗细菌内毒素作用,可在由IL-1α、IL-6和ET-1介导的机体炎症等病理损伤过程中发挥一定的药理作用。 The aim of the research was to investigate the anti-inflammatory mechanism of Pulsatillae Decoction(PD), and the levels of interleukin (IL) -1 α, interleukin-6 (IL-6) and endothelin-1 ( ET-1 ) secreted by cultured rat intestinal microvaseular endothelial cells were determined after treatment with its active ingredients, namely anemoside 134, anemonin, berberine ,jatrorrhizine, palmatine, aesculin, and esculetin. Endothelial cell was challenged with 1 μg· mL-1 lipopolysaccharide (LPS)for 3 h, and then treated respectively with PD at three concentrations (1,5 and 10 mg. mL-1 )and its seven ingredients at three concentrations (1,5 and 10 μg· mL^-1 ). The cells were in- cubated at 37 ℃ in a cell incubator for 21 h. The supernatants were collected and the content of IL-1α,IL-6 and ET-1 analyzed by ELISA kits. The results showed that as compared with LPS control group ,the IL-hx contents were significantly decreased in the high- dose group of PD ,the middle- and high-dose groups of anemoside B4 ,the middle- and high-dose groups of berberine and the high-dose group of palmatine; The IL-6 contents were significantly decreased in middle- and high-dose groups of PD, the high-dose group of anemonin,the middle- and high-dose groups of aesculin and the high-dose group of esculetin;The ET-1 contents were significantly decreased in the middle-dose group of PD, the middle- and high-dose groups of anemonin and the middle- and high-dose groups of esculetin. It suggested that PD may exert beneficial therapeutic actions on systemic inflammatory response syndrome, muhiple organ dysfunction syndrome and immune diseases mediated by IL-1α,IL-6 and ETA.
