肺炎支原体感染对BALB/c小鼠肺组织半胱氨酰白三烯及其受体表达的影响 被引量:3

摘要 目的探讨肺炎支原体(MP)感染对BALB/c小鼠肺组织半胱氨酰白三烯(CysLTs)及其受体(CysLTs-R)表达的影响。方法100只BALB/c小鼠随机分成肺炎支原体感染组(MP组)和PBS对照组(N组),每组50只,分别于接种后3 d、7 d、14 d、21 d和30 d从各组中取5只小鼠,取其肺组织,收集肺泡灌流液(BALF),用ELISA试剂盒检测CysLTs的含量;同时将肺脏固定于中性甲醛中,制作病理切片,HE染色观察病理组织学变化;免疫组织化学技术检测CysLTs-R的表达情况;用Western blot实验检测肺组织中CysLTs-R的表达变化。结果病理组织学观察结果显示,MP感染BALB/c小鼠3 d后,即出现间质性炎症改变,7 d后炎症明显,14 d后炎症逐渐减轻并于30 d时基本消退;ELISA检测结果表明,与N组比较,MP组BALF中CysLTs含量明显增高(P<0.05),于感染后7 d和14 d达到高峰,而后呈下降趋势;免疫组化结果显示,在MP感染后,CysLTs-R蛋白在气管上皮细胞表面的表达增强;Western blot结果显示MP组小鼠于感染后3 d、7 d、14 d、21 d和30 d,肺组织中CysLTs-R蛋白的表达量均高于N组。结论 MP感染可以显著增加正常BALB/c小鼠中CysLTs及CysLTs-R的表达水平。 Objective To investigate the expression of leukotriene and its receptor in BALB/c mice infected by mycoplasma pneumonia. Methods A total of 100 BALB/c mice were randomly divided into mycoplasma pneumoniae (MP) infection group (MP group ) and PBS control group (N group) (n =50). Five mice were sacrificed from each group after the vaccination 3 d, 7 d, 14 d, 21 d and 30 d, respectively, and lung tissue was col- lected for harvest of alveolar perfusate (BALF). CysLTs content was determined using ELISA kit; the rest of the lung tissue was fixed in formalin for sections, HE staining was performed for the observation of histological changes; CysLTs-R expression was detected by both say and western blot analysis. Results As shown in histopathological observation results, interstitial inflammatory changes occurred 3 d after MP in- fection, significantly increased on day 7, then gradually decreased after day 14, and was basically subsided on day 30; ELISA test results show that compared with normal group, MP CysLTs in BALF were significantly increased (P 〈 0.05 ), which peaked at day 7 and day 14 and then declined ; immunohistochemistry analysis showed that CysLTs-R protein expression on the cell surface was enhanced in tracheal epithelial after MP infection; Western blot analysis showed lung tissue CysLTs-R protein expression levels higher after infection at all tested time points than the N group. Conclu- sion MP infection can significantly increase the CysLTs and CysLTs-R expression in normal BALB / e mice.
