摘要
目的 研究IL 4和IL 10对CD4+T细胞上CXC趋化性细胞因子受体 4(CXCR4)表达的调控。方法 采用流式细胞术 ,实时定量逆转录PCR(RT PCR) ,细胞因子的趋化性分析以及Scatchard分析法检测CXCR4和CXCR4mR NA。结果 IL 4和IL 10能够显著上调或下调CD4+T细胞上CXCR4的表达。SDF 1诱导的CD4+T细胞趋化性同样也受到IL 4和IL 10的调控。IL 4和IL 10能分别上调或下调CD4+T细胞中CXCR4的mRNA表达。在新鲜分离的CD4+T细胞上只存在一种亲和力的CXCR4(Kd6 3nmol·L-1)。而IL 4刺激的CD4+T细胞存在两种不同亲和力的CXCR4(Kd14 4nmol·L-1和Kd2 14 6nmol·L-1)。IL 4和IL 10通过环腺苷酸和环鸟苷酸双重信号途径对CD4+T细胞上CXCR4表达的调控与Ca2 +激活刺激无关。IL 4和IL 10可能是通过CD2 6来调控CXCR4在CD4+T细胞上的表达。结论 IL 4和IL 10对CXCR4 SDF 1受体 配体对的调节 。
Objective To investigate the regulation of CXCR4 expression on CD4 + T lymphocytes by IL 4 and IL 10. Methods CXCR4 and CXCR4 mRNA was detected by flow cytometry, real time quantitative reverse transcription(RT PCR), chemotacxis assay,and scatchard analysis. Results IL 4 and IL 10 significantly up or down regulated CXCR4 expression on CD4 + T lymphocytes, respectively. SDF 1 induced CD4 + T lymphocytes chemotaxis was also regulated by IL 4 and IL 10, correspondingly. IL 4 and IL 10 up or down regulated CXCR4 mRNA expression on CD4 + T lymphocytes, respectively. A kind of CXCR4 with affinity ( K d 6 3 nmol·L -1 ) on flesh isolated CD4 + T lymphocytes, and two kinds of CXCR4 with different affinities ( K d1 4 4 nmol·L -1 and K d2 14 6 nmol·L -1 ) on IL 4 stimulated CD4 + T lymphocytes. IL 4 and IL 10 regulate CXCR4 expression on CD4 + T lymphocytes via both cAMP and cGMP signaling pathways. The capacity of IL 4 and IL 10 to regulate CXCR4 on CD4 + T lymphocytes was not linked to calcium mobilization stimulation. The effect of IL 4 and IL 10 may be via CD26 molecule to regulate CXCR4 expression on CD4 + Tlymphocytes. ConclutionThe effects of IL 4 and IL 10 on CXCR4 SDF 1 recept ligand pair may be especially important in cytekine/chemokine environment concerning the inflammatory processes and the progression of HIV infection.
