摘要
为了更好地降低生物学实验成本,对市场上质粒提取试剂盒中吸附柱可重复利用性进行了初步研究。探讨了不同溶液、保存时间、pH值、离子浓度以及温度对吸附柱再吸附能力的影响。结果表明:使用后的吸附柱经过一定的处理可以有效地恢复其对质粒DNA的再吸附能力。确定最佳处理条件为:在离子浓度为0.01mol·L-1、pH值为2.04的PBS缓冲溶液中于4℃保存6d。经过处理,吸附柱的再吸附能力相对其第1次吸附能力有所增强。
In order to reduce the cost of biology experiment,we studied preliminarily the reuse of adsorption column for the plasmid kit on the market.The effects of different solution,retention time,pH value,ion concentration and temperature on recovering the adsorbability of adsorption column were discussed.The results indicated that the used adsorption column's adsorbability was recovered effectively by definite handling methods.The optimum handling conditions were as follows:the adsorption column was preserved in the PBS buffer with pH value of 2.04 and ion concentration of 0.01 mol · L-1 for 6 d at 4 ℃.The adsorbability of the handled adsorption column was enhanced to some degree compared with its first adsorption.
