摘要
本研究采用 SDS- PAGE法 ,系统研究了苎麻脱胶高效菌株 T85 - 2 6 0纯培养过程中的胞外酶系 ,初步得出如下结论 :能在不同的培养基中组成型产生 41种胞外蛋白 (含胞外酶或亚基 ) ,分子量为 1 0 .0 KD- 1 2 0 .0 KD;可能能分泌分子量分别为 6 0 .3KD、49.5 KD、33.5 KD、2 9.5 KD的 4种甘露聚糖酶 ,分子量为 2 8.6 KD的果胶酶 ,较高浓度的镁离子有利于果胶酶的产生 ,分子量分别为 72 .9KD、35 .5 KD、31 .6 KD、1 9.9KD的 4种蛋白酶 ;T85 - 2 6 0嗜好甘露糖。
The extracellular enzymes from highly efficiently degumming bacterium T85-260 for ramie under pure culture were systematically studied by SDS-PAGE. Conclusions run as follows:41 extracellular proteins (including enzymes and subunits) with M.W. ranging from 10.0 KD to 120.0KD could be secreted under different media; 4 kinds of mannanases with M.W.60.3KD, 49.5KD, 33.5KD, 29.5KD, one pectinase weighted 28.6KD, 4 proteases with M.W.72.9KD, 33.5KD, 31.6KD, 19.9KD could be secreted. Mannose is a good C-source for T85-260. The above results should be testified by ramie degumming process.
