摘要
目的探讨Livin阳性乳腺癌中细胞周期调控相关蛋白的变化,以初步明确Livin对细胞周期调控相关蛋白的影响。方法采用免疫组化SP法检测Livin在45例乳腺癌中的表达,利用Western blot法检测45例乳腺癌中Cyclin D、Cyclin A、Cyclin E的表达。结果 45例乳腺癌中Livin阳性率为75.56%(34/45)。Livin阳性乳腺癌中,Cyclin A的阳性率为88.23%(30/34),而在Livin阴性乳腺癌中,Cyclin A的阳性率为54.55%(6/11),差异有统计学意义(P<0.05);Livin阳性乳腺癌中,Cyclin D的阳性率为61.76%(21/34),而Livin阴性乳腺癌中,Cyclin D的阳性率为81.82%(9/11),差异无统计学意义(P>0.05);在Livin阳性乳腺癌中,Cyclin E的阳性率为82.35%(28/34),而在Livin阴性乳腺癌中,Cyclin E的阳性率为45.46%(5/11),差异有统计学意义(P<0.05)。乳腺癌组织中Livin与Cyclin A、Cyclin E蛋白表达显著相关(P<0.05)。结论 Livin蛋白在乳腺癌中表达上调,提示其可能在促进乳腺癌发生、发展中起重要作用。Livin表达可导致Cyclin A和Cyclin E表达上调,而对Cyclin D表达作用不明显,提示Livin除了抗细胞凋亡作用外,还可能通过引起细胞周期调控紊乱促进乳腺癌的发生、发展。
Purpose To investigate the changes of cell cycle control-related proteins in Livin-positive breast carcinoma and preliminarily definitize the effects of Livin on the cell cycle control-related proteins in Livin-positive breast carcinoma.Methods Livin was detected by immunohistochemical analysis (IHC,SP method)in 45 breast cancerous tissues.Cyclin D,Cyclin A,Cyclin E were detected in 45 cases of breast cancerous tissues by Western blot.Results Among the 45 breast carcinoma,the positive rate of Livin expression was 75.56% (34/45).In the Livin-positive breast carcinoma,the positive rate of Cyclin A was 88.23% (30/34).But in the Livinnegative breast carcinoma,the positive rate of Cyclin A was 54.55% (6/11).There was a significant difference between the two groups (P <0.05).The positive rate of Cyclin D in the Livin-positive breast carcinoma was 61.76% (21/34),which lower than that in the Livin-negative breast carcinoma (81.82%,9/11),but the difference between the two groups was not statistically significant (P > 0.05).Expression of Cyclin E in the Livin-positive breast carcinoma was 82.35% (28/34),however,in the Livin-negative breast carcinoma,the positive rate of Cyclin E was 45.46% (5/11).There was significant difference between the two groups (P < 0.05).The protein expression of Livin with Cyclin A and Cyclin E were correlated separately (P < 0.05).Conclusions The upregulated expression of Livin is caually linked to the development and progression of the breast carcinoma.The expression of Cyclin A and Cyclin E in Livin-positive breast carcinoma are upregulated,however,the expression of Cyclin D has no dramatic changes.The results show that the disorder of cell cycle control induced by Livin play an important role in the development of breast carcinoma.
