PCR直接测序法与ARMS检测非小细胞肺癌EGFR基因突变 被引量：5

摘要 目的 比较PCR直接测序法与蝎形探针扩增阻滞突变系统（amplification refractory mutation system,ARMS）检测非小细胞肺癌（non-small cell lung cancer,NSCLC）中EGFR基因突变的敏感性.方法 用直接测序发与ARMS法同时检测154例NSCLC中EGFR基因第18、19、20、21外显子的突变情况.结果 男性84例,女性70例,年龄33 ～ 86岁,手术切除标本71例、活检标本79例,其中鳞状细胞癌27例、腺癌121例、腺鳞癌2例;另4例为胸水.测序法32例突变（32/154,20.8％）,ARMS法52例突变（53/154,33.8％）.两种方法均检测到突变者29例,均无突变者99例,直接测序法检测到突变而ARMS法阴性者3例,反之23例,不一致率差异有显著性（P＜0.000 1）.结论 检测NSCLC中EGFR突变情况时,ARMS法较直接测序法操作步骤少,突变率高.
