用出生4天的SD大鼠睾丸作器官块组织培养,观察不同浓度的咖啡因单独、与腺苷或苯异丙基腺苷协同对睾丸培养块的生殖细胞数量及所有细胞成份DNA合成的影响。结果表明,高浓度的咖啡因(0.6mM)引起培养块的生殖细胞数目及有DNA合成标记的细胞核数减少。高剂量的腺苷(0.3及0.6mM)能抵抗同剂量的咖啡因对生殖细胞数量的影响;而低10倍浓度的腺苷及苯异丙基腺苷则无此作用。但加入以上两种药物都能拮抗高浓度咖啡因所导致睾丸培养块DNA合成的下降。本文讨论了咖啡因及腺苷作用的可能性机制。
4-day-old rat testicular organ explant cultures were used to investigate the effects of caffeine alone and in combination with adenosine or(-)-N^6 (phenylisopropyl)adenosine(L-PIA)on germ cell development and DNA synthesizing activity.At a dose of 0.6mM,caffeine decreased the numbers of germ cells as well as the overall DNA synthesis rate in the organ explants.When caffeine was coadministered with adenosine,only high equimolar doses(0.3 and 0.6mM)of ad enosine could ameliorate the detrimental effect of caffeine on the germ cells,while tenfold lower doses of adenosine or L-PIA could not.Regarding the overall DNA sythesis in the testicular explants,application of adenosine or L-PIA togerther with a high dose of caffeine counteracted caffeine effects.The mechanisms of the effects with caffeine and adenosine have been discussed.
