食蟹猴源食道口线虫和缩小特尼登线虫的形态学鉴定及基于其ITS2基因的分子种系发育分析 被引量:2

摘要 对采集自食蟹猴肠道血结节中的3种线虫进行形态观察后鉴定,并用PCR扩增其ITS2基因后进行序列测定分析。结果显示,3种线虫分别为尖形食道口线虫、双叉食道口线虫及缩小特尼登线虫。测序结果表明,3种虫体ITS2基因序列的大小均为216bp。邻位连接法构建的种系发育树显示,本研究中测定的尖形食道口线虫、双叉食道口线虫和缩小特尼登线虫序列在进化树上自成独立的拓扑分支。以上结果为人类食道口线虫病及缩小特尼登线虫病建立分子鉴别诊断方法提供了分子标记。 Three species of nematodes from Macaca fascicularis were identified according to morpho- logy. The ITS2 rDNA sequences of the nematodes were amplified by PCR, sequenced and analyzed. In result, the nematodes were Oesophagostomurn aculeatum , 0. bi furcum and Ternidens deminutus. The sequencing result indicated that all the ITS2 rDNA sequences were 216 bp in size. Phylogenetic trees constructed using neighbor-joining method showed that the sequenced O. aculeatum, O. bifurcum and T. deminutus were situated in separated topology branches, respectively. These results provide available molecular marker to establish molecular differential diagnosis for human diseases caused by O. aculeatum, 0. bi furcum and T. deminutus.
