摘要
为探讨外膜蛋白AⅡ(OMPAⅡ)作为维氏气单胞菌共同保护性抗原的可能性,对不同宿主来源的维氏气单胞菌OMPAⅡ基因进行了克隆,获得了1 100bp左右的目的基因,并利用多种生物学软件对其序列进行了比较。结果表明,不同宿主来源的维氏气单胞菌OMPAⅡ基因序列的同源性在95.1%以上,编码氨基酸序列的同源性在97.3%以上,而且所有分析序列均具有明显的保守结构域,说明不同宿主来源的维氏气单胞菌OMPAⅡ具有很高的保守性;相对于ATCC35624和YP004393583.1来说,它们之间的差异主要表现在,CVCC3700、QXF、WL161、NJ34、NN725和BAF63175.1的OMPAⅡ基因缺失了3个碱基(CAA)。本研究结果为OMPAⅡ作为维氏气单胞菌基因工程亚单位疫苗候选成分提供了一定的理论依据。
To investigate the possibility of outer membrane protein AⅡ (OMPAⅡ) as a common pro- tective antigen of Aeromonas veronii ,A. veronii OMPAⅡ genes about 1 100 bp from different hosts were obtained and the comparison and analysis were carried out between them by using several bio-software. The homology of these A. veronii OMPAⅡ genes were higher than 95. 1% at the nucleotide level and higher than 97.3% at the amino acid level. And the same conserved domains were found,which showed that A. veronii OMPAⅡfrom different hosts had high-degree conservation. Difference between them was mainly manifested in that OMPAⅡ genes of CVCC3700 ,QXF,WL161 ,NJ34 ,NN725 and BAF63175.1 had the deletion of 3 bases(CAA). This showed that OMPA Ⅱ might be considered as an important candidate for genetic engineering subunit vaccines against A. veronii.
