摘要
以黄海黄杆菌 YS- 94 12 - 130菌株为出发株 ,经亚硝基胍、硫酸二乙酯、紫外线 ( UV)与微波( MI)复合诱变和自然选育 ,获得 1株产低温碱性蛋白酶高产稳定的突变株 YS- 94 12 - 130 - SW1- 10 4 ,其产低温碱性蛋白酶为出发株的 16倍。在摇瓶发酵培养条件下 ,酶活力达 2 32 0 U/ ml( 2 0℃测定 ) ,该诱变株具有 Refr、Strr、Ampr和 Met-、Lys-标记。突变株与出发菌株在细胞形态上比较 。
In screening high producer of low temperature alkaline protease, F. yellowsea YS 9412 130 is chosen as original strain, which is treated repeatedly with various mutagens(NTG、DEM、UV and Microwave). The mutant YS 9412 130 SW 1 104 with high and stable yield of alkaline protease is selected after repeated natural selection. The alkaline protease activity of this mutant is in the 16 fold of the original strain, reaching 2 320U/ml (assayed 20℃). The mutant has marked with Ref r、Str r、Amp r 、Met - and Lys -.The result shows that there are no significant differences between the cells of the mutant and the original strain.
