非洲菊叶片愈伤组织诱导和不定芽分化的研究 被引量:4

摘要 以非洲菊3个品种叶片为材料,研究了不同基因型、叶片叶龄和叶片不同部位以及激素对非洲菊愈伤组织诱导和不定芽分化的影响。结果表明:诱导愈伤组织较为适宜的外植体为嫩叶,以MS+6-BA 1.0~5.0 mg/L+NAA 0.2 mg/L为培养基,愈伤组织诱导率均>98%;诱导分化不定芽较为适宜的外植体为叶柄,最适培养基为MS+TDZ 2.0 mg/L+NAA 0.2 mg/L,但不定芽分化率较低,仅为4.2%。 The effects of different genotypes,leaf ages,parts and hormones on callus induction and bud differentiation were studied with leaf explants .The results showed that induction rate was all above 98%when tender leaves were the explants of callus induction and culture medium was MS +6-BA 1.0-5.0 mg/L+NAA 0.2 mg/L .The callus from petiole explants produced adventitious bud by subculture on MS+TDZ 2.0 mg/L+NAA 0.2 mg/L,which had a low differentiation rate of 4.2%.
