摘要
干扰素γ诱导蛋白30(interferon,gamma-inducible protein 30,IFI30)在MHC-Ⅱ限制性抗原加工递呈过程和MHC-Ⅰ限制性抗原交叉递呈过程中起关键性作用。利用3个不同浓度PRRSV感染猪肺支气管巨噬细胞(PAM),应用qPCR在不同时间点检测IFI30基因表达水平。结果表明:0.08,0.10,0.12 MOI 3个剂量PRRSV感染PAM,IFI30基因表达差异不显著(P>0.05);PRRSV感染1.5 h IFI30表达量降低,但差异不显著(P>0.05),之后表达量上调;在感染后12,24,36 h IFI30基因表达量极显著提高(P<0.01),之后IFI30基因表达量有所下降(P<0.05)。IFI30基因在PRRSV感染PAM过程中表达量的变化,可能是PRRSV与宿主相互作用的机制之一。
Interferon,gamma-inducible protein 30 (IFI30)plays a key role in MHC classⅡ-restricted antigen processing and MHCⅠ-restricted cross-presentation .In our study, PAMs were infected by PRRSV of three different concentrations,and the levels of IFI30 mRNA expression of different time were detected by qPCR .The results showed that the IFI30 expression differences were not significant when PAMs were infected by three different concentrations,0.08 MOI,0.1 0 MOI and 0.1 2 MOI(P&gt;0.05), respectively .The expression of IFI30 infected by PRRSV after 1.5 h was reduced,but the difference was not significant (P&gt;0.05 ),and then the expression of IFI30 was increased,the expression difference was very significant when PAMs were infected by PRRSV after 1 2 h,24 h and 36 h (P&lt;0.01 ),but then the expression of IFI30 was reduced (P&lt;0.05 ).The results indicated that the IFI30 expression change in the process of PRRSV infecting PAM may be one of the mechanisms which interact between PRRSV and host .
