摘要
采用室内培养实验方法 ,研究了不同浓度和不同时间胺草磷 (APM )对蚕豆根系细胞有丝分裂及根系蛋白质和POD同工酶的影响。结果表明 :(1)不同浓度APM处理后 ,可明显提高有丝分裂指数 ,但与APM浓度的增加无明显线性关系。(2)5μmol/L浓度处理的根系 ,其蛋白质谱带出现增加与缺失现象 ,与对照相比 ,增加了4条谱带 ,在组蛋白区域缺失1条谱带。(3)根系内POD同工酶的变化趋势与蛋白质变化基本吻合 ,1~4μmol/L处理未呈现明显变化 ,但在5μmol/L下与对照相比诱导出一条活性很强的POD3 谱带。综上所述 ,5μmol/LAPM浓度处理可以认为是蚕豆根尖中一些生化性状产生变化的临界浓度。
In this paper, the effects of various concentration (1~5 μmol/L) with various treatment times (8 h,10 h and 12h) of APM on mitosis, protein and POD isozyme of root cell were investigated in Vicia faba. The results indicated that: (1) After treatment, the mitosis index was obviously increased, the promoting effects on mitosis of prophase and metaphase cell were more significant, although there was no notable linear relationship between the increase of mitosis index and the changes of APM concentration. (2) Compared with control test, some protein bands appeared, while the other band disappeared in the root treated with 5 μmol/L of APM. The molecular weights of 4 appearing bands were: 28 KD, 66 KD, 78 KD and 88 KD, respectively. The disappearing band was a 14 KD protein in histone area. There was no notable changes of protein in root treated with 1~4 μmol/L of APM. (3)The trend of isozyme changes was similar to that of protein. An obvious changes occurred with 1~4 μmol/L treatment tests, but 5 μmol/L of APM made POD increase significantly. Compared with control group, a high activity POD band was induced. Above all, 5 μmol/L of APM was believed to be the concentration threshold on the occurrence of some biochemical changes of root cell in Vicia faba.
