补肾活血颗粒对帕金森大鼠脑腺苷A2A受体影响的研究 被引量:3

摘要 目的探讨补肾活血颗粒治疗帕金森病的作用机制。方法用6-羟基多巴损毁脑右侧内侧前脑束的方法建立帕金森大鼠模型。将33只SD模型大鼠随机分为治疗组18只和对照组15只,另设正常组15只,治疗组以中药补肾活血颗粒灌胃每日1次,连续8周。对照组及正常组同法以生理盐水灌胃8周,灌胃结束、观察帕金森大鼠旋转行为变化,后断头取脑,免疫荧光法观察大鼠脑腺苷A2A受体变化。结果治疗组大鼠旋转行为较对照组改善,治疗组大鼠脑纹状体A2A受体表达较对照组明显减弱(P<0.01),模型组比正常组表达显著增强(P<0.01)。三组大鼠黑质中A2A受体均无表达。结论补肾活血颗粒能改善帕金森大鼠旋转行为,抑制大鼠纹状体腺苷A2A受体表达,对帕金森病治疗起到较好疗效。 Objective To explore the mechanism of treating Parkinson's disease(PD)with Bushen Huoxue Granule(BSHXG). Methods Establishing rat model of PD with 6-hydroxydopamine lesion as me- dial forebrain bundle the right side of the brain of. 33 PD rats modes were randomly divided into treatment group 18 (BSHXG group) and control group 15. Additionally other 15 SD rats were selected in normal group. BSHXG was applied for treatment group once daily for 8 weeks. Physiologic saline was applied for control group and normal group in the same way. Garage end, observed changes in the rotational behavior of PD rats. After decapitated, Immunofluorescence was used to observe the changes of adenosine A2A re- ceptor in rat brain. Results Rotational behavior of rats in the treatment group was much better improved than in the control group. A2A receptor expression in the treatment group compared with the control group was significantly decreased( P 〈 0.01 ). A2A receptor expression in the model group was significantly in- creased than the normal group(P 〈0. 01 ). A2A receptor were not expression in three groups of rats sub- stantia nigra. ConcLusion BSHXG can improve the rotational behavior of PD rats, inhibit adenosine A2A receptor expression in PD rat striatum, and has a significant therapeutic effect on the treatment of Parkinson's disease.
