加味过敏煎对哮喘小鼠气道炎症的抑制作用 被引量:7

摘要 【目的】探讨加味过敏煎对哮喘小鼠气道炎症的影响。【方法】选用BALB/c雌性小鼠60只,按随机数字表法分为6组,分别为空白对照组(空白组),模型对照组(模型组),地塞米松阳性对照组(地塞米松组,剂量为1.25 mg·kg-1·d-1),加味过敏煎高、中、低剂量组(中药高、中、低剂量组,剂量分别为27.2、13.6、6.8 g·kg-1·d-1),每组10只。采用卵白蛋白(OVA)致敏、雾化方法复制小鼠哮喘模型。各组于末次激发24 h后,取外周血进行白细胞总数及分类计数;右肺组织切片,苏木精—伊红(HE)染色观察各组肺组织病理学改变并进行评分。【结果】与正常对照组比较,模型组外周血中白细胞总数、淋巴细胞、嗜酸性粒细胞、中性粒细胞均显著升高(P<0.05或P<0.01),肺组织病变程度严重(P<0.01)。与模型组比较,中药高、中剂量组白细胞总数、嗜酸粒细胞数量显著减少(P<0.05或P<0.01)。中药高剂量组的淋巴细胞、中性粒细胞显著减少,差异均有统计学意义(P<0.05)。HE染色显示:中药高、中剂量组能使肺组织中细支气管、血管周围的嗜酸性粒细胞浸润得到控制(P<0.01),减轻支气管周围炎性病变程度。【结论】加味过敏煎对哮喘小鼠气道炎症具有一定抑制作用。 Objective To explore the impact of modified Guomin Decoction, an experienced herbal prescription, on airway inflammation in mice with asthma. Methods Sixty female BALB/c mice were randomly divided into 6 groups: blank control group, model control group, dexamethasone positive control group (dexamethasone in the dosage of 1.25 mg.kg-l.d-1) and high-, middle- and low-dose modified Guomin Decoction groups (in the dosage of 27.2, 13.6, 6.8 g.kg-1.d-1, respectively), 10 mice in each group. The method of ovalbumin-induced sensitization following by challenge with ovalbumin atomization was adopted to establish asthma mouse model. Twenty-four hours after the last challenge, peripheral blood was sampled for the total count and classification of white blood cells. The right lung tissue sections were stained with hematoxilin and eosin (HE) for the observation of pathological changes and for pathological scoring. Results Compared with the blank control group, the total number of peripheral white blood cells, and peripheral lymphocytes, eosinophilie granulocytes and neutrophile granulocytes counts were significantly increased in peripheral blood (P〈0.05 or P 〈0.01), and pathological lesions in mice lung tissue was severe (P〈0.01) in the model group. Compared with the model group, the total number of white blood cells and eosinophilic granulocytes count were decreased in high- and middle-dose modified Guomin Decoction groups (P〈O.05 or P〈0.01) . The number of lymphocytes and neutrophile granulocytes in high-dose modified Guomin Decoction group was obviously fewer than that in model group (P〈0.05) . HE staining results revealed that eosinophilic infiltration around bronchioles and vessels of lung tissue was controlled (P〈0.01, compared with that in the model group), and inflammation around bronchia was alleviated in high- and middle- dose modified Guomin Decoction groups. Conclusion Modified Guomin Decoction has inhibitory effect on airway inflammation in mice with asthma.
