盆腔炎颗粒对慢性盆腔炎大鼠NF-κB及相关指标表达的影响

Effect of Penqiangyan Granule on Expression of NF-κB and Related Indexes of Rats with Chronic Pelvic Inflammatory

目的:研究盆腔炎颗粒对慢性盆腔炎大鼠核因子κB(NF-κB)及相关指标表达的影响。方法:以混合细菌加机械损伤法复制大鼠慢性盆腔炎模型,随机分为正常对照组、假手术组、模型组、盆炎净组及盆腔炎颗粒大、中、小剂量组,造模2周后,各治疗组灌胃给药2周,免疫组化法检测各组大鼠子宫组织中NF-κB、核因子κB结合蛋白(IκB)、细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子-α(TNF-α)的表达,逆转录-聚合酶链反应法检测各组大鼠子宫组织中ICAM-1mRNA、TNF-αmRNA表达。结果:盆腔炎颗粒可以显著降低模型大鼠NF-κB、IκB、TNF-α、ICAM-1、TNF-αmRNA及ICAM-1mRNA的表达,盆腔炎中剂量组子宫组织NF-κB与TNF-αmRNA及ICAM-1mRNA表达之间均呈显著正相关。结论:NF-κB活化的抑制在活血补肾法治疗慢性盆腔炎抗炎及抗粘连机制中起重要作用。

Objective：To study the eitect oI Penqiangyan granule（PQYG） on the expression of NF-kB and related indexes of rats with chronic pelvic inflammatory（CPI）. Methods：The models of CPI were established with mixed bacteria and mechanical injury,and were randomly divided into normal group,sham operation group,model group,Penyanjing group,and PQYG group which consisted of large,medium and low doses. Af- ter 2 weeks the model established successfully,medicines were intragastrically administrated to CPI rats for 2 weeks,expressions of NF-KB,IKB,ICAM-1,TNF-α were tested with inmunohistoehemical method,expres- sions of ICAM-lmRNA and TNF-αmRNA were detected by reverse transcriptase polymerase chain reaction. Results：PQYG can significantly reduce the expressions of NF-KB,IKB,TNF-α,ICAM-1,TNF-αmRNA and I- CAM-lmRNA. NF-KB had significant positive correlation with TNF-ctmRNA and ICAM-lmRNA in medi- um-dose PQYG group. Conclusion：Inhibiting NF-KB activation is the mechanism of Huoxue Bushen method in treating CPI through anti-inflammatory and anti-adhesion.
