维生素E拮抗绿脓菌素抑菌作用的实验研究

摘要 目的通过观察维生素E干预金黄色葡萄球菌的绿脓菌素药敏试验揭示维生素E拮抗绿脓菌素抑菌的作用机制。方法收集从2011年6月15日~2012年12月15日期间,临床微生物标本中分离到的铜绿假单胞菌50株分别编号1~50号和金黄色葡萄球菌3株,分别编号A,B,C。铜绿假单胞菌经肉汤培养基增菌后提取绿脓菌素,绿脓菌素与相对应的铜绿假单胞菌编号=致,制备绿脓菌素纸片,每株金黄色葡萄球菌配制菌悬液,分别用相同编号、相同含量的2个绿脓菌素药敏纸片做药敏试验,M—H平板上涂布维生素E的作为试验组,其不涂维生素E的作为相对应的对照组。结果金黄色葡萄球菌A,B,C各株绿脓菌素纸片抑菌环直径用平均值和标准差(-X±s)表示分别为B实验=16.39±5.12mm,A实验=19.38±4.38mm,C实验=21.04±4.72mm,对照组分别为B对M=19.33±5.61mm,A对M=21.30±4.88mm,C时照=23.18±5.92mm,试验组抑菌环明显小于对照组。数据经SPSSl7.0软件进行成对样本t检验统计-分剐为tA=4.2402,tc=5.2661,tB=8.1531,P值均〈0.01。结论维生素E通过清除绿脓菌素抑菌过程中产生的自由基起到桔抗其抑菌的作用。 Objective To reveal Pyocyanin vitamin E antagonize Pyocyanin inhibitory mechanism of action by observing the vitamin E treatment of Staphylococcus aureus susceptibility testing. Metheds Collected from June 15,2011 to December 15 2012 clinical microbiology specimens isolated from Pseudomonas aeruginosa 50 No. l-50,respectively, and Staphylococcus a ureus ,No3, numbered A,B,C and P. aeruginosa broth enrichment by extraction after pyocyanin pyocyanine with the corre- sponding serial number of P. aeruginosa was prepared pyocyanine paper,Staphylococcus aureus per plant cocci bacterial sus pension prepared, respectively,with the same number, the same content in two pyocyanin susceptibility paper susceptibility testing to do,MH plates coated withvitamin E as a test group,which was not coated with vltamin E as the crrespondittg the control group. Results Staphylococcus aureus A,B and C each strain Pyocyanin disk zone diameters with a mean and stand- ard deviation (-x±s) were expressed as B test= 16.39±5.12 mm,A test=19.38±4.38 mm,C test=21.04±t4.72 mm,the control group was B control= 19.33±5.61 mm,A control=21.30±4.88 mm,C control=23.18±5.92 ram,inhibition zone test group was significantly less than the control group. Conclusion Vitamin E Pyocyanin by clearing the free radicals produced during inhibition plays antagonize the antibacterial effect.
