摘要
目的探讨自拟柴青解郁汤在抑郁症小鼠的抗抑郁作用。方法将50只昆明种雄性小鼠随机分为5组:对照组、模型组、盐酸氟西汀组、柴青解郁汤组、柴青解郁汤与盐酸氟西汀联合组(联合用药组),每组10只。采用慢性轻度不可预见性应激加孤养复制抑郁症小鼠模型,观察各组小鼠体质量变化、糖水消耗实验、悬尾实验及强迫游泳实验结果并进行比较。结果实验前(0 d)各组小鼠体质量及糖水偏爱百分比比较差异均无统计学意义(P>0.05);造模后(14 d),模型组、盐酸氟西汀组、柴青解郁汤组和联合用药组小鼠体质量及糖水偏爱百分比显著低于对照组,差异均有统计学意义(P<0.05);但模型组、盐酸氟西汀组、柴青解郁汤组和联合用药组之间小鼠体质量及糖水偏爱百分比比较差异均无统计学意义(P>0.05)。给药后(35 d)盐酸氟西汀组、柴青解郁汤组和联合用药组小鼠体质量及糖水偏爱百分比显著高于模型组,差异均有统计学意义(P<0.05);联合用药组小鼠体质量及糖水偏爱百分比显著高于盐酸氟西汀组和柴青解郁汤组,差异均有统计学意义(P<0.05);盐酸氟西汀组和柴青解郁汤组小鼠体质量及糖水偏爱百分比比较差异无统计学意义(P>0.05)。模型组小鼠游泳不动时间和悬尾不动时间显著长于对照组,差异均有统计学意义(P<0.01);盐酸氟西汀组、柴青解郁汤组、联合用药组小鼠游泳不动时间和悬尾不动时间显著短于模型组,差异有统计学意义(P<0.05,P<0.01);联合用药组小鼠游泳不动时间和悬尾不动时间显著短于盐酸氟西汀组和柴青解郁汤组,差异均有统计学意义(P<0.05)。结论自拟柴青解郁汤具有抗抑郁作用,且与盐酸氟西汀联合用药抗抑郁效果更佳。
Objective To investigate the antidepressant effects of self-made Chaiqing Jieyutang on depressive disorder in mice. Methods Fifty Kunming male mice were randomly divided into five groups : control group, model group, fluoxetine group, Chaiqing Jieyutang group, Chaiqing Jieyutang and fluoxetine combination group ( combined medication group ) , 10 mice in each group. The depressive disorder mice were made by chronic mild unpredictable stress and separate feeding. The changes of body weight and the results of sugar consumption experiment, tail suspension test and forced swimming test were observed and compared among the five groups. Results There was no significant difference in the body weight and sucrose preference percentage of mice among the five groups before experiment (0 day) (P 〉 0. 05 ). The body weight and sucrose preference percentage of mice in model group, fluoxetine group, Chaiqing Jieyutang group, combined medication group were significantly lower than those in control group after modeling( 14 days) (P 〈 0.05 ). There was no significant difference in the body weight and sucrose preference percentage of mice among model group, fluoxetine group, Chaiqing Jieyutang group and combined medication group after modeling( 14 days ) ( P 〉 0.05 ). The body weight and sucrose preference percentage of mice in fluoxetine group, Chaiqing Jieyutang group and combined medication group were significantly higher than those in model group after mod- eling(35 days)(P 〈0.05 ). The body weight and sucrose preference percentage of mice in combined medication group were significantly higher than those in fluoxetine group and Chaiqing Jieyutang group after modeling( 35 days ) (P 〈 0.05 ). There was no significant difference in the body weight and sucrose preference percentage of mice between fluoxetine group and Chaiqing Jieyutang group after modeling ( 35 days) ( P 〉 0.05 ). The immobility time of mice in tail suspension test and forced swimming test in model group was significantly longer than that in control group(P 〈 0. 01 ). The immobility time of mice in tail suspension test and forced swimming test in fluoxetine group, Chaiqing Jieyutang group, combined medication group were significantly shorter than those in model group( P 〈 0.05, P 〈 0.01 ). The immobility time of mice in tail suspension test and forced swimming test in combined medication group were significantly shorter than those in fluoxetine group and Chaiqing Jieyutang group (P 〈 0.05). Conclusion Self-made Chaiqing dieyutang has antidepressant effects. Chaiqing Jieyutang combination with fluoxertine hydrochloride has better antidepressant effects.
