窄谱中波紫外线对银屑病患者单核细胞趋化蛋白-1及其受体CCR2 mRNA表达的影响 被引量:7

Effects and significance of narrowband-ultraviolet B phototherapy on the expression of MCP-1 and its receptor CCR2 mRNA in patients with psoriasis vulgaris

摘要 目的:探讨窄谱中波紫外线(NB-UVB)对银屑病患者单核细胞趋化蛋白(monocyte chemotactic protein,MCP)-1及其受体CCR2 mRNA表达水平的影响。方法:采用双抗体夹心酶联免疫吸附法(ELISA)分别检测30例银屑病患者NB-UVB照射治疗前、后外周血CC型趋化因子MCP-1的表达量。采用反转录-聚合酶链反应(RT-PCR)法检测NB-UVB照射治疗前、后患者外周血单一核细胞CCR2 mRNA的表达水平。结果:寻常性银屑病患者治疗前外周血中MCP-1、CCR2 mRNA表达量明显高于正常对照组(P_1<0.01,P_2<0.01),NB-UVB照射治疗显著改善病情后,患者外周血中MCP-1、CCR2 mRNA表达量较治疗前明显降低,差异具有统计学意义(P_1<0.01,P_2<0.01)。结论:MCP-1及其受体CCR2可能与银屑病的发病相关,NB-UVB可能通过降低银屑病患者外周血MCP-1及其受体CCR2的表达而发挥作用。 Objective: To observe the effects and significance of narrowband-ultraviolet B(NB-UVB) phototherapy on the expression of MCP-1 and its receptor CCR2 mRNA in patients with psoriasis vulgaris. Methods: Thirty patients with psoriasis vulgaris were enrolled. They were treated with NB-UVB. Serum levels of MCP-1 were detected with enzyme-linked immunosorbent assay(ELISA) before and after treatment. Reverse transcription-polymerase chain reaction(RT-PCR) was applied to semi-quantitatively analyze the CCR2 mRNA expression in the peripheral blood mononuclear cells (PBMC) from 30 pa- tients with psoriasis vulgaris before and after treatment, and 30 healthy controls. Results: The expression levels of MCP-1, CCR2 mRNA in patients with psoriasis vulgaris before treatment were significantly higher than those of the healthy controls (PI〈0.01,P2〈0.01). The MCP-1,CCR2 mRNA levels of therapeutic group after treatment were significantly lower than the levels before treatment(Pl〈0.01,P2〈O.01). Conclusion: MCP-1 and its receptor CCR2 may be involved in the pathogenesis of psoriasis vulgaris, and the efficacy of NB-UVB phototherapy in the treatment of psoriasis may be partially due to decreased expression of MCP-1 and its receptor CCR2.
