Expression of estrogen receptors in peripheral blood mononuclear cells from female patients with systemic lupus erythematosus

摘要 目的:检测外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)中雌激素受体在基因和蛋白水平的表达,分析其表达类型与女性系统性红斑狼疮的相关性。方法:收集46例女性SLE患者和33例健康体检者空腹静脉血,分离PBMCs,采用RT-PCR法检测PBMCs中雌激素受体(estrogen receptor,ER)αmRNA、ERβmRNA的表达,荧光免疫技术检测ERα和ERβ在蛋白水平的表达。分析两组ER表达的差异。结果:SLE患者组PBMCs中ER亚型在基因和蛋白水平上的表达率与正常对照组无显著差异(P>0.05)。结论:ER的表达类型与女性SLE没有相关性。 Objective:Examine the messenger RNA(mRNA) and protein expressions of estrogen receptors(ER) in peripheral blood mononuclear cells(PBMCs) of female patients with systemic lupus erythematosus(SLE) and analyze the correlations between ER and SLE.Methods:The fasting venous blood was drawn from 48 female patients with SLE and 33 healthy women,and PBMCs were isolated.The expressions of ERα mRNA and ERβ mRNA in PBMCs were measured by reverse transcription polymerase chain reaction,and the protein expressions of ERα and ERβ were measured by immunofluorescence.And the differences between ER and SLE were analyzed.Results:The mRNA and protein expression ratio of ER subtype had no significant difference between the female patients with SLE and healthy control ones(P>0.05).Conclusion:There is no correlation between the expression types of ERs and female patients with SLE.
