摘要 目的:探讨Eag1钾离子通道、缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)与血管内皮生长因子(vascular endothelial growth factor,VEGF)在不同病变宫颈组织中的表达及其临床意义.方法:应用免疫组化方法(S-P法)检测40例慢性宫颈炎、40例CIN和100例宫颈癌组织中Eag1、HIF-1α及VEGF表达情况,并分析其与宫颈癌临床病理因素的关系.结果:宫颈鳞癌组织内Eag1、HIF-1α与VEGF蛋白的表达明显高于宫颈上皮内瘤变组和慢性炎症组(P<0.01);非角化型宫颈鳞癌组织中Eag1、HIF-1α与VEGF的表达高于角化型宫颈鳞癌(均为P<0.05); Ⅱb期及以上期宫颈癌组织中Eag1、HIF-1α与VEGF 表达高于Ⅰ~Ⅱa期(均为P<0.05);伴有淋巴结转移者三者的表达均高于无淋巴结转移(均为P<0.05);放疗前三者的表达高于放疗后(均为P<0.05);Eag1、HIF-1α与VEGF三种蛋白在宫颈鳞癌组织表达呈正相关(均为P<0.05).结论:Eag1、HIF-1α和VEGF的表达在宫颈鳞癌发生发展过程中起重要作用,与宫颈鳞癌进展密切相关,是宫颈鳞癌早期诊断和治疗的潜在靶点. Objective:Explore the expression and its clinical significance of Eag1 potassium channel,hypoxia inducible factor-1α(HIF-1α) and vascular endothelial growth factor(VEGF) in different cervical lesion tissues.Methods:The expression of Eag1,VEGF and HIF-1α were measured in 40 cases of chronic cervicitis,40 cases of cervical intraepithelial neoplasia and 100 cases of cervical cancer by SP immunohistochemiscal method.The relationship between clinical pathological factors and cervical cancer was discussed.Results:Immunohistochemistry showed that the expressions of Eag1,HIF-1α and VEGF protein in cervical squamous cell carcinoma tissues were significantly higher than the chronic cervicitis group and the cervical intraepithelial neoplasia group(P<0.01).The expressions of Eag1,HIF-1α and VEGF in non-keratinizing squamous cell carcinoma were higher than the keratinizing squamous cell carcinoma group(P<0.05).The expressions of Eag1,HIF-1α and VEGF in the stage of Ⅱb and above were higher thanⅠ~Ⅱa period(P<0.05).These three protein expressions in the lymph node metastasis of cervical squamous cell carcinoma were higher than which without it(P<0.05).These three protein expressions before radiotherapy in cervical squamous cell carcinoma were higher than the post-radiotherapy group(P<0.05).The expressions of Eag1,HIF-1α and VEGF in cervical squamous cell carcinoma were positively correlated.Conclusion:Eag1,HIF-1α and VEGF protein expressions play important roles in the development of cervical carcinoma.And they are closely related with each other.Eag1,HIF-1α and VEGF can be used to predict prognosis in cervical squamous cell carcinoma.
