摘要
目的:探讨Wnt/β-连环蛋白(Wnt/β-catenin)在高糖诱导的新生大鼠心肌成纤维细胞CFs增殖中的作用。方法:分离新生乳鼠CFs,构建高糖(25 mmol/L)诱导CFs增殖细胞模型。采用Wnt/β-catenin抑制剂XAV939以及激动剂SKL2001调控Wnt/β-catenin信号通路。MTT法和细胞计数检测心肌细胞增殖,Western blot法检测大鼠CFs中β-catenin的表达,实时荧光定量PCR(qPCR)检测增殖细胞核抗原(PCNA)的表达。结果:25 mmol/L高糖刺激48 h后CFs的细胞增殖显著增加,β-catenin表达显著升高(P<0.01),XAV939能抑制CFs增殖(P<0.01),降低高糖诱导β-catenin及PCNA的表达(P<0.01),SKL2001能对抗XAV939抑制高糖诱导CFs增殖的作用。结论:高糖诱导的CFs增殖可能与激活Wnt/β-catenin途径有关。
Objective: To investigate the role of Wnt/ β-catenin in neonatal rat cardiac fibroblast proliferation induced by high glucose. Methods: Neonatal rat cardiac fibroblasts were isolated, and high glucose (25mmol/L) induced cardiac fibroblast proliferation model was established; Wnt/β-cate- nin inhibitor XAV939 and agonist SKL2001 was used to regulate Wnt/ [3-catenin signaling pathway. Cell proliferation was detected by MTY assay and cell count. Western blot was used to detect the ex- pression of β-catenin. The expression of proliferating cell nucleus antigen (PCNA) was assayed by qPCR. Results: After incubating with 25 mmol/L high glucose for 48 h, cardiac fibroblasts prolifera- ted obviously, and the expression of β-catenin was increased significantly XAV939 inhibited cardiac fibroblast proliferation (P 〈 0.01 ) and reduced in fibroblasts ( P 〈 0.01 ) the expression of β-cateninand PCNA (P 〈 0.01 ) ; SKL2001 could resist the inhibiting effect of XAV939 in cardiac fibroblast proliferation. Conclusions: Proliferation of cardiac fibroblasts induced by high glucose may be associ- ated with activation of Wnt / β-catenin pathway.
